摘  要:本试验旨在优化艾蒿多糖(AAP)的提取工艺,并研究其组成成分和体外抗氧化活性。在单因素试验的基础上,以AAP提取率作为响应值,通过响应面分析得出AAP最佳提取工艺;分析纯化后AAP的形状结构和单糖组成及分子质量;并通过测定1,1-二苯基-2-三硝基苯肼自由基(DPPH·)、羟自由基(·OH)、超氧阴离子自由基(O_(2)^(-)·)的清除率以及铁还原力,评估AAP的抗氧化活性。结果表明:1)通过响应面优化AAP的最佳提取工艺为液料比35∶1、酶解时间4 h、温度50℃,AAP提取率为12.46%,可溶性多糖含量为297.11 mg/g。2)纯化后AAP呈现出表面粗糙的立方体状颗粒、长链片状、圆球状等多种不规则块状形态;含阿拉伯糖、半乳糖、葡萄糖、木糖、甘露糖5种单糖,其分子质量比为1.68∶3.54∶80.81∶2.38∶11.59;平均分子质量为114.786 ku,属于吡喃糖。3)相同浓度下,AAP组DPPH·清除率和铁还原率在0.05~1.00 mg/mL浓度下均极显著低于阳性对照组(P<0.01),·OH清除率在0.40~1.00 mg/mL浓度下和O_(2)^(-)·清除率在0.20~1.00 mg/mL浓度下也极显著低于阳性对照组(P<0.01)。AAP对DPPH·、·OH和O_(2)^(-)·清除率的半数抑制浓度(IC_(50))分别为0.0552、0.8684和0.8392 mg/mL。综上所述,本研究优化了传统AAP提取工艺,并通过组分分析及体外抗氧化活性测定,发现AAP具有较强的抗氧化活性,为AAP作为饲料添加剂的开发与利用提供坚实的理论依据。This experiment was conducted to optimize the extraction process of Artemisia argyi polysaccharides(AAP),and investigated it components and in vitro antioxidant activity.Based on single-factor experiments,the extraction yield of AAP was taken as the response value,and the optimal extraction conditions were determined through response surface methodology.The shape,structure,monosaccharide composition and molecular weight of the purified AAP were analyzed.Additionally,its antioxidant activity of AAP was evaluated by measuring the scavenging rates of 1,1-diphenyl-2-picrylhydrazyl radicals(DPPH·),hydroxyl radicals(·OH)and superoxide anion radicals(O_(2)^(-)·)as well as its iron reducing power.The results showed as follows:1)the best extraction process of AAP was optimized by response surface with liquid to material ratio of 35∶1,enzymatic hydrolysis time of 4 h and temperature of 50℃.The extraction rate of AAP was 12.46%,and the content of soluble polysaccharides was 297.11 mg/g.2)The purified AAP exhibited a variety of irregular particle shapes,including rough cubic particles,long-chain flakes and spherical forms.It contained five monosaccharides:arabinose,galactose,glucose,xylose and mannose,with molecular weight ratios of 1.68∶3.54∶80.81∶2.38∶11.59.The average molecular weight was 114.786 ku,and it was classified as a pyranose.3)At the same concentration,the DPPH·scavenging rate and iron reduction rate at concentrations ranging from 0.05 to 1.0 mg/mL in the AAP group were significantly lower than those in the positive control group(P<0.01),and the·OH scavenging rate at concentrations ranging from 0.40 to 1.00 mg/mL and the O-2·scavenging rate at concentrations ranging from 0.20 to 1.00 mg/mL were also significantly lower than those in the positive control group(P<0.01).The half-maximal inhibitory concentrations(IC_(50))for DPPH·,·OH,and O_(2)^(-)·scavenging rates were 0.0552,0.8684 and 0.8392 mg/mL,respectively.In conclusion,this study optimizes the traditional AAP extraction process and demon
