摘　　要：目的探讨miR-770-5p、着丝粒蛋白抗体结合因子(ZWINT)在肺癌组织中的表达及临床意义。方法本研究为回顾性队列研究。采用目的抽样法选取2020年3月至2023年11月中国人民解放军空军第986医院收治的115例经病理组织学检查确诊的肺癌患者临床资料。采用实时荧光定量聚合酶链反应检测癌组织与癌旁组织中miR-770-5p、ZWINT表达,比较不同临床特征患者miR-770-5p、ZWINT差异;根据不同miR-770-5p、ZWINT表达程度分为miR-770-5p高表达组(n=65)、miR-770-5p低表达组(n=50),ZWINT高表达组(n=69)、ZWINT低表达组(n=46),比较不同组别患者预后情况。根据患者预后不同,分为预后良好组(n=83)和预后不良组(n=32)。比较不同预后患者miR-770-5p、ZWINT表达差异。采用Cox回归分析影响肺癌患者预后的因素,同期绘制受试者操作特征(ROC)曲线分析miR-770-5p、ZWINT表达及联合对肺癌患者预后的预测价值;采用双变量Pearson相关性检验miR-770-5p、ZWINT表达量之间的相关性;采用Spearman秩相关分析检验及点二列相关性检验miR-770-5p、ZWINT表达量与临床特征之间的相关性。结果115例肺癌患者中男71例,女44例;年龄(61.91±3.28)岁,年龄范围为57~67岁。肺癌组织中miR-770-5p表达量低于癌旁组织[(0.61±0.04)比(1.03±0.10)],ZWINT表达量高于癌旁组织[(0.48±0.11)比(0.21±0.09)],差异均有统计学意义(t值分别为43.08、20.26,均P<0.05)。不同TNM分期、分化程度、淋巴结转移患者miR-770-5p、ZWINT表达量比较,差异均有统计学意义(均P<0.05);不同性别、病理类型、肿瘤直径、癌胚抗原、糖类抗原199、癌胚抗原125情况患者的miR-770-5p、ZWINT表达量比较,差异均无统计学意义(均P>0.05)。miR-770-5p表达量与TNM分期、分化程度、淋巴结转移情况均呈负相关(均r<0,均P<0.05),ZWINT表达量与TNM分期、分化程度、淋巴结转移情况均呈正相关(均r>0,均P<0.05)。miR-770-5p高表达组总生存率为ObjectiveTo investigate the expressions of miR-770-5p and Zeste White 10-interacting kinetochore protein(ZWINT)in lung cancer tissues and their clinical significances.MethodsThis was a retrospective cohort study involving 115 pathologically confirmed lung cancer cases admitted in the 986th Hospital of PLAAF from March 2020 to November 2023 by objective sampling.Real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to detect the expression levels of miR-770-5p and ZWINT in lung cancer tissues and adjacent tissues,and their expression levels were detected in lung cancer patients with different clinical characteristics.According to different levels of miR-770-5p and ZWINT,lung cancer patients were divided into high miR-770-5p expression group(n=65),low miR-770-5p expression group(n=50),high ZWINT expression group(n=69),and low ZWINT expression group(n=46).Prognosis was compared among the four groups.According to the different outcomes of prognosis,they were divided into a good prognosis group(n=83)and a poor prognosis group(n=32).Expression levels of miR-770-5p and ZWINT between the good and poor prognosis groups were compared.Cox regression analysis was used to identify risk factors for the prognosis of lung cancer,and receiver operating characteristic(ROC)curve was drawn to analyze the predictive value of miR-770-5p,ZWINT,and their combination in the prognosis of lung cancer.Bivariate Pearson correlation test was used to examine the correlation between miR-770-5p and ZWINT expression levels.Spearman rank correlation analysis and point-biserial correlation were used to examine the correlation between miR-770-5p,ZWINT expression and clinical characteristics.ResultsOut of 115 lung cancer patients,there were 71 males and 44 females,with a mean age of 61.91±3.28(57-67)years old.The expression level of miR-770-5p in lung cancer tissues was significantly lower than that in adjacent tissues(0.61±0.04 vs1.03±0.10),and the expression level of ZWINT was significantly higher than that in adjacent t
