摘 要:目的:该研究旨在从母乳喂养新生儿粪便中筛选分离出对幽门螺杆菌具有拮抗作用的益生菌菌株,并通过体外试验评价其安全性和益生性。方法:采用牛津杯琼脂扩散法进行幽门螺杆菌抑制实验,分别通过溶血性、药敏特性和模拟胃肠液耐受性、抗氧化能力实验评价益生菌菌株安全性和益生性。结果:从健康母乳喂养新生儿粪便中分离筛选出20株益生菌菌株。20株菌株中,XF-35对幽门螺杆菌抑菌圈直径最大,显著高于其它菌株和对照菌株LGG(P<0.05)。结合分子生物学鉴定结果分析,将菌株XF-35鉴定为副格氏乳杆菌。副格氏乳杆菌XF-35菌株无溶血性,与LGG相比,其表现出相似的抗生素耐药性模式。同时,副格氏乳杆菌XF-35表现出较高的人工模拟胃肠液耐受性和抗氧化能力。结论:副格氏乳杆菌XF-35是一株具有良好安全性和益生性,并能拮抗幽门螺杆菌的益生菌菌株,具有作为益生菌预防幽门螺杆菌感染的潜力。Objective:This study aimed to screen and isolate probiotic strains from the feces of breastfed neonates that could antagonize Helicobacter pylori,and evaluate their safety and probiotic characteristics via in vitro experiments.Methods:The oxford cup agar diffusion method was used to conduct the experiment of inhibition on H.pylori,and the safety and probiotic properties were evaluated through experiments of hemolysis,antibiotic sensitivity,tolerance to artificial simulated gastrointestinal juices and antioxidant capacity,respectively.Results:A total of 20 probiotic strains were successfully isolated and screened from the feces of healthy breastfed neonates.Among them,strain XF-35 had the largest inhibition zone diameter against H.pylori,which was markedly greater than that of the other strains and the control strain LGG(P<0.05).Based on molecular biological identification,strain XF-35 was identified as Lactobacillus paragasseri.Notably,Lactobacillus paragasseri XF-35 strain demonstrated no hemolytic activity and shared a similar antibiotic resistance pattern with LGG.In addition,this strain displayed a high level of tolerance to artificial simulated gastrointestinal juices as well as robust antioxidant capacity.Conclusion:Lactobacillus paragasseri XF-35 is a probiotic strain with dependable safety and favorable probiotic properties,capable of exerting an antagonistic effect against Helicobacter pylori.It holds the potential to serve as a promising probiotic candidate for preventing Helicobacter pylori infection.
