摘  要:目的探讨家兔肝、脑、脊髓组织中4种RNA相对表达量与死亡时间(PMI)的关系。方法选取家兔26只,溺死后将尸体置于恒温恒湿(温度25℃、湿度50%)环境中,选择死后0~72 h内8个时间点提取肝、脑、脊髓组织RNA,利用实时荧光定量聚合酶链式反应(PCR)技术检测ATP合成酶-H+转运V0结构域a1亚基蛋白(ATP6 V0A1)信使RNA(mRNA)、二肽基肽酶4(DPP4)mRNA、调节突触膜胞吐作用蛋白2(RIMS2)环状RNA(circRNA)、迪格弗梅尔基-奥尔克劳森综合征蛋白(DYM)circRNA 4种RNA,使用SPSS 25.0统计软件对目标RNA相对表达量进行统计分析,并对目标RNA与PMI进行曲线回归分析。结果随着PMI延长,ATP6 V0A1 mRNA在家兔肝脏内的相对表达量明显高于脑和脊髓组织;DPP4 mRNA在家兔肝脏内的相对表达量在死后8 h开始升高,死后24 h达到峰值后开始急剧下降,但仍明显高于脑和脊髓组织;相比之下,RIMS2 circRNA、DYM circRNA 2种circRNA在肝、脑和脊髓组织中的相对表达量呈平稳缓慢下降趋势,其中DYM circRNA在3种组织中的相对表达量变化趋势最稳定。结论circRNA的表达水平与PMI的相关性较好,联合应用其他分子标记,在法医死亡时间推断及侦查破案过程中具有一定的应用前景。Objective To investigate the relationship between post-mortem interval(PMI)and the relative expression levels of four RNAs in liver,brain and spinal cord of rabbits.Methods Twenty-six domestic rabbits were selected and euthanized by drowning.Their carcasses were then placed in an environment with constant temperature(25℃)and humidity(50%).RNA was extracted from liver,brain,and spinal cord tissues at eight time points within 0 to 72 hours post-mortem.Real-time fluorescent quantitative polymerase chain reaction(PCR)technology was utilized to detect four types of RNA:ATP6 V0A1 mRNA,DPP4 mRNA,RIMS2 circRNA,and DYM circRNA.SPSS 25.0 statistical software was employed to conduct statistical analysis on the relative expression levels of the target RNAs.Additionally,curve regression analysis was performed between the target RNAs and PMI.Results As PMI increased,the relative expression level of ATP6 V0A1 mRNA in the liver of rabbits was significantly higher than that in the brain and spinal cord.The relative expression level of DPP4 mRNA in the liver began to increase at 8 hours after death,reached a peak at 24 hours,and then decreased sharply,but was still significantly higher than that in the brain and spinal cord.In comparison,the relative expression levels of the two circRNAs,RIMS2 circRNA and DYM circRNA,exhibited a trend of steady and gradual decline in liver,brain,and spinal cord tissues.Among them,the relative expression level of DYM circRNA demonstrated the most stable trend across all three tissue types.Conclusion The expression level of circRNA has a good correlation with PMI.Combined with other molecular markers,it has certain application prospects in forensic PMI estimation and criminal investigation.
