摘 要:目的 研究羽扇豆醇对人卵巢癌SKOV3细胞增殖、迁移及侵袭的影响。方法 以人卵巢癌SKOV3细胞为研究对象,根据加入羽扇豆醇浓度不同分为四组:对照组(0μmol/L)、低剂量组(20μmol/L)、中剂量组(40μmol/L)、高剂量组(80μmol/L);用MTT法检测细胞的增殖能力;用划痕实验检测细胞的迁移能力;用Transwell法检测细胞的侵袭能力;用Western blot检测PCNA、ASAP1蛋白的表达。结果 与对照组相比较,不同剂量组羽扇豆醇干预后的人卵巢癌SKOV3细胞增殖、迁移及侵袭能力均明显下降(P<0.05),且对羽扇豆醇呈剂量依赖性(P<0.05);与对照组相比较,不同剂量组羽扇豆醇干预后的人卵巢癌SKOV3细胞中PCNA、ASAP1蛋白的表达量均明显下降(P<0.05),且对羽扇豆醇呈剂量依赖性(P<0.05)。结论 羽扇豆醇可抑制人卵巢癌SKOV3细胞株的增殖、迁移及侵袭能力,其作用的机制可能通过下调PCNA与ASAP1蛋白的表达有关。Objective To investigate the effects of lupeol on the proliferation,migration,and invasion of human ovarian cancer SKOV3 cells.Methods Human ovarian cancer SKOV3 cells were used as the research object and divided into four groups according to the different concentrations of added lupeol which were control group(0 μmol/L),low-dose group(20 μmol/L),medium dose group(40 μmol/L),and high-dose group(80 μmol/L).Detected the proliferation ability of cells using MTT assay.Use scratch test to detect the migration ability of cells.Use Transwell method to detect the invasive ability of cells.Use Western blot to detect the expression of PCNA and ASAP1 proteins.Results Compared with the control group,the proliferation,migration,and invasion abilities of human ovarian cancer SKOV3 cells were significantly reduced after intervention with different doses of lupeol(P<0.05),and showed a dose-dependent effect on lupeol(P<0.05).Compared with the control group,the expression levels of PCNA and ASAP1 proteins in SKOV3 ovarian cancer cells after intervention with different doses of lupeol were significantly decreased(P<0.05),and showed a dose-dependent effect on lupeol(P<0.05).Conclusion lupeol can inhibit the proliferation,migration,and invasion ability of human ovarian cancer SKOV3 cell line,and its mechanism of action may be related to the downregulation of PCNA and ASAP1 protein expression.
