摘 要:目的:探讨不同规格(HB、CK、SK)葛藤花的有效成分含量及抗氧化活性差异。方法:紫外分光光度法检测样品总黄酮含量,高效液相色谱(HPLC)法测定葛藤花中染料木素、鸢尾苷和染料木苷3种成分含量,比色法检测葛藤花甲醇提取液的Fe^(3+)还原能力、DPPH自由基清除能力、ABTS自由基清除能力和总抗氧化能力(FRAP)。通过SPSS 26.0软件对数据进行主成分分析、聚类分析和相关性分析。结果:3种规格样品中总黄酮、染料木苷、鸢尾苷和染料木素含量分别为31.25~42.09 mg/g、0.91~1.12 mg/g、21.38~28.19 mg/g和0.33~0.58 mg/g。Fe^(3+)还原能力表现为SK>CK>HB,DPPH自由基清除能力表现为CK>SK>HB,ABTS自由基清除能力和FRAP的规律性不明显。CK规格的葛藤花综合排名靠前,品质最佳,聚类分析结果与形态分类一致。结论:不同规格葛藤花品质差异明显,CK规格综合品质最好。本研究为葛藤花药材的资源开发和花类药材的产地分级加工提供了一定参考。Objective:To explore the differences in the content of active ingredients and antioxidant activities among different specifications(HB,CK,SK)of Puerariae omeoensis Flos.Methods:The contents of total flavonoids,genistin,tectoridin and genistein in the samples were detected by ultraviolet spectrophotometry(UV)and high-performance liquid chromatography(HPLC).The Fe^(3+)reducing ability,DPPH free radical scavenging ability,ABTS free radical scavenging ability and ferric ion reducing antioxidant power(FRAP)were detected by colorimetry.Principal component analysis,cluster analysis,and correlation analysis were performed by software of SPSS 26.0.Results:The contents of total flavonoids,genistin,tectoridin and genistein in the three specifications ranged from 31.25 to 42.09 mg/g,0.91 to 1.12 mg/g,21.38 to 28.19 mg/g,and 0.33 to 0.58 mg/g,respectively.The Fe^(3+)reducing ability was in the order of SK>CK>HB,while the DPPH radical scavenging ability was CK>SK>HB.The patterns of ABTS radical scavenging ability and FRAP were not obvious.The CK specification with the highest ranking showed the best quality,and the clustering analysis result were consistent with the morphological classification.Conclusion:There were significant differences in the quality of Puerariae omeoensis Flos of different specifications,with the CK specification having the best overall quality.This study provided a certain reference for the resource development of Puerariae omeoensis Flos and the classification and processing of floral medicinal materials.
