摘 要:目的:研究蒙药胡日查六味丸抗偏头痛的作用机制。方法:通过查阅相关文献和应用中药系统药理学数据库与分析平台(TCMSP)、ETCM、GeneCards、TTD等数据库,筛选出蒙药胡日查六味丸的有效成分、作用靶点及偏头痛的疾病靶点;进一步以Venny 2.1.0交集出蒙药胡日查六味丸抗偏头痛作用靶点,将以上潜在靶点通过STRING平台建立蛋白质-蛋白质相互作用(PPI)网络图,通过David数据库进行基因本体论(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析;应用Cytoscape 3.8.2软件构建蒙药胡日查六味丸的活性成分-靶点-信号通路网络图,筛选主要有效成分、核心靶点及信号通路。结果:网络药理学分析结果显示,蒙药胡日查六味丸抗偏头痛的核心活性成分为山柰酚、槲皮素、异鼠李素、木犀草素、Bicyclo[3.1.1]heptan-2-one,6,6-dimethyl-(1R)-(6,6-二甲基-(1R)双环[3.1.1]-2-庚酮)等,核心靶点为丝氨酸/苏氨酸蛋白激酶1(Akt Serine/Threonine Kinase 1,AKT1)、表皮生长因子受体(Epidermal Growth Factor Receptor,EGFR)、酪氨酸蛋白激酶(Sarcoma,SRC)、信号转导因子和转录激活因子(Signal Transducer and Activator of Transcription,STAT)3、磷脂酰肌醇-3-激酶催化亚基α(Phosphatidylinositol-3-Kinase Catalytic Subunit Alpha,PIK3CA)等,主要通路为神经活性配体受体相互作用、癌症途径、钙离子信号通路、环磷酸腺苷信号通路、化学致癌-受体活化途径、5-羟色胺能突触等。结论:本研究通过网络药理学技术系统探讨蒙药胡日查六味丸治疗偏头痛的作用机制,为该药的进一步应用与研究提供参考。Objective:To study the mechanism of Mongolian medicine Hurizha Liuwei pills(胡日查六味丸)in treating migraine.Methods:By consulting relevant literature and applying Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP,ETCM(The Encyclopedia of Traditional Chinese Medicine),GeneCards,TTD(Therapeutic Target Database)and other databases,the effective ingredients of Mongolian medicine Hurizha Liuwei pills,action targets and disease targets of migraine were screened out.Furthermore,Venny 2.1.0 intersection was used to identify the target of the anti-migraine effect of Mongolian medicine Hurizha Liuwei pills,and the protein-protein interaction(PPI)network diagram was established through the STRING platform.The Gene Ontology(GO)enrichment analysis was conducted through the David database and the pathway annotation analysis from the Kyoto Encyclopedia of Genes and Genomes(KEGG)was conducted.The active ingredient-target-signal pathway network diagram of Mongolian medicine Hurizha Liuwei pills was constructed by Cytoscape 3.8.2 software,and the main effective ingredients,core targets and signal pathways were screened.Results:Network pharmacological analysis showed that the core active ingredients of Mongolian medicine Hurizha Liuwei Pill in treating migraine were kaempferol,quercetin,isorhamnetin and luteolin,Bicyclo[3.1.1]heptan-2-one,6,6-dimethyl-(1R)-(6,6-dimethyl-(1R)bicyclo[3.1.1]-2-heptanone),etc.The core targets were AKT1,EGFR,SRC,STAT3,PIK3CA,etc.The main pathways were neuroactive ligand receptor interaction,cancer pathway,calcium ion signaling pathway,cyclic adenosine phosphate signaling pathway,chemical carcinogenic-receptor activation pathway,5-hydroxytryptaminergic synapse,etc.Conclusion:This study systematically explores the mechanism of Mongolian medicine Huricha Liuwei pills in treating migraine through network pharmacology technology,aiming to provide reference for the further application and research of this drug.
