摘 要:短萼黄连是黄连的变种,为国家二级保护野生植物。为了深入了解野生短萼黄连种子的萌发特性,实现种子活力的快速测定,选用5因素4水平的正交试验,分析了浸种时间、浸种温度、TTC质量浓度、染色时间、染色温度等5个条件对染色效果的影响,并筛选出最优染色条件。结果表明:各因素对生活力的影响从大到小排序为浸种时间、染色时间、TTC质量浓度、染色温度、浸种温度,浸种时间和染色时间对生活力的测定具有显著影响;短萼黄连种子TTC染色的最佳条件为20℃浸种24h后用10.0 g·L^(-1)的TTC溶液35℃避光染色24 h,此时生活力测定值为50.00%,略高于发芽率,能够反映种子生活力的真实情况。TTC染色法能快速、准确、有效地测定短萼黄连种子的生活力,在短萼黄连种子质量检测中具有重要的应用价值。Coptis chinensis var.brevisepala is a variant of Coptis chinensis,and is a national secondary protected wild plant.In order to deeply understand the seed germination characteristics of wild Coptis chinensis var.brevisepala and realize the rapid determination of seed vigor,the effects of soaking time,soaking temperature,TTC concentration,dyeing time and dyeing temperature on the TTC dyeing effect were analyzed by orthogonal test with 5 factors and 4 levels,and the optimal dyeing conditions were selected.The results showed that the order of the effects of various factors on seed viability was soaking time,dyeing time,TTC concentration,dyeing temperature and soaking temperature.Soaking time and dyeing time had significant effects on the determination of seed viability.The optimum condition for TTC staining was that the seeds were soaked at 20℃for 24 h and then stained with 10.0 g·L^(-1)TTC solution at 35℃for 24 h.At this time,the measured value of seed viability was 50.00%,which was slightly higher than the germination rate and could reflect the real situation of seed viability.TTC method can determine the seed viability of Coptis chinensis var.brevisepala quickly,accurately,and effectively,and has important application value in seed quality detection.
