摘 要:试验旨在研究沙棘黄酮(Sea-buckthorn flavone,SF)对黄曲霉毒素B1(aflatoxin B1,AFB1)诱发的鸡肝细胞损伤的保护作用。通过灌流法获得鸡原代肝细胞,分为对照组(Ⅰ)、沙棘黄酮组(Ⅱ)、模型组(Ⅲ)、试验组(Ⅳ)。对照组(Ⅰ组:未添加沙棘黄酮),加入含有10%胎牛血清的DMEM培养基;Ⅱ组添加140μg·mL^(-1)沙棘黄酮;Ⅲ组添加0.05μg·mL^(-1)黄曲霉毒素B1;Ⅳ组添加0.05μg·mL^(-1)黄曲霉毒素B1+140μg·mL^(-1)沙棘黄酮。在对各组肝细胞处理12 h后,采用MTT法测定细胞存活率,采用试剂盒测定细胞培养液中转氨酶活性及抗氧化指标。结果显示:与对照组相比,Ⅲ组肝细胞培养液的ALT和AST活性均显著升高(P<0.05),细胞存活率极显著降低(P<0.01);与Ⅲ组相比,Ⅳ组肝细胞培养液中ALT和AST活性显著下降(P<0.05),细胞存活率极显著升高(P<0.01);与对照组相比,Ⅲ组肝细胞中GSH-Px和SOD活性极显著降低(P<0.01),MDA含量极显著提高(P<0.01);与Ⅲ组相比,Ⅳ组肝细胞中GSH-Px和SOD活性极显著升高(P<0.01),MDA含量极显著降低(P<0.01)。结果表明:沙棘黄酮对AFB1诱发的鸡肝细胞损伤具有保护作用。The protective effect of sea-buckthorn flavone(SF)on chicken liver cell injury induced by aflatoxin B1(AFB1)was investigated in this study.Chicken primary hepatocytes were obtained by perfusion method and were divided into four group,including the control group(Ⅰ),seabuckthorn flavonoids group(Ⅱ),the model group(Ⅲ),and the test group(Ⅳ).Control cells without added drugs(DMEM culture medium containing 10%fetal bovine serum was used);groupⅡwith cells treated with 140μg·mL^(-1) SF was added;groupⅢwith cells treated with 0.05μg·mL^(-1) AFB1 was added;groupⅣ(0.05μg·mL^(-1) AFB1+140μg·mL^(-1) SF).After 12 h of treatment,the cell viabi-lity was determined by MTT assay,the transaminase activity in the cell culture medium was measured,and the antioxidant indicators were determined.The results showed that compared with the control group,the ALT and AST activities in the groupⅢcell culture medium were significantly increased(P<0.05),and the cell viability was signi-ficantly decreased(P<0.01);compared with the groupⅢ,the ALT and AST activities in the groupⅣcell culture medium were significantly decreased(P<0.05),and the cell viability was significantly increased(P<0.01);compared decrea with the control group,the GSH-Px and SOD activities in the groupⅢliver cells were significantly decreased(P<0.01),and the MDA content was significantly increased(P<0.01);compared with groupⅢ,the GSH-Px and SOD activities were markedly increased(P<0.01),and the MDA content was significantly decreased(P<0.01)in groupⅣ.The results showed that sea-buckthorn flavone had a protective effect on AFB1 induced chicken liver cell injury.
