摘  要:优化减肥食品中蒽醌苷元类物质的酸水解提取方法,并采用高效液相色谱法进行测定。以番泻苷元A、番泻苷元B、芦荟大黄素、大黄素、大黄酸、大黄酚、大黄素甲醚、橙黄决明素为对照品,以蒽醌苷元类物质得率为考察指标,在单因素实验基础上,采用响应面法对酸水解提取蒽醌苷元类成分得率影响较大的因素(硫酸质量分数、酸水解温度、超声辅助提取酸水解时间)进行优化,结果表明,酸水解提取蒽醌苷元类物质最优条件为甲醇沸水浴回流提取45min,硫酸质量分数为15%,酸水解温度为75℃,超声辅助提取酸水解时间为50 min。The acid hydrolysis extraction process of anthraquinoneaglycones in weight loss foods was optimized,and determination was performed using high performance liguid chromatography(HPLC).Using sennosides A and B,aloe-emodin,emodin,rhein,chrysophanol,physcion,aurantio-obtusin as reference standards,and taking the yield of anthraquinoneaglycones as the evaluation index,response surface methodology(RSM)was employed to optimize the significant factors affecting the yield of anthraquinoneaglycone components in acid hydrolysis extraction based on single-factor experiments.These factors included sulfuric acid mass fraction,acid hydrolysis temperature,and ultrasound-assisted extraction time for acid hydrolysis.The results showed that the optimal conditions for acid hydrolysis extraction of anthraquinoneaglycones were as follows:reflux extraction with methanol boiling water bath for 45 min,sulfuric acid mass fraction of 15%,acid hydrolysis temperature of 75℃,and ultrasonic-assisted extraction and acid hydrolysis time of 50 min.
