膀胱癌患者癌组织WWOX、WBP2表达及临床预后意义

摘　　要：目的探讨膀胱癌患者癌组织含WW域的氧化还原酶(WWOX)、WW域结合蛋白2(WBP2)表达及临床预后意义。方法选取2017年12月至2019年12月驻马店市中心医院收治的112例膀胱癌患者,采用免疫组化法检测癌组织(膀胱癌组)及对应癌旁组织(癌旁组)WWOX、WBP2水平。采用χ^(2)检验探讨膀胱癌患者癌组织WWOX、WBP2表达与其病理特征的关系;采用多因素Cox回归分析探讨预后因素。结果膀胱癌组WBP2阳性率高于癌旁组,WWOX阳性率低于癌旁组(P<0.05)。低分化、肿瘤直径≥1 cm、TNM分期为Ⅲ~Ⅳ期、淋巴结转移、血管间隙浸润患者WBP2阳性率、WWOX阴性率分别高于中/高分化、肿瘤直径<1 cm、TNM分期为Ⅰ~Ⅱ期、无淋巴结转移、无血管间隙浸润患者(P<0.05)。WWOX阴性表达患者3 a生存率高于WWOX阳性表达患者(P<0.05);WBP2阳性表达患者3 a生存率低于WBP2阴性表达患者(P<0.05)。Ⅲ~Ⅳ期、淋巴结转移、WWOX阴性、WBP2阳性是膀胱癌患者预后危险因素(P<0.05)。结论WWOX在膀胱癌组织中主要呈阴性表达,WBP2主要呈阳性表达,二者均与膀胱癌患者临床病理特征密切相关,有望作为评估膀胱癌预后的标志物。Objective To explore the expression and clinical prognostic significance of WW domain-containing oxidoreductase(WWOX)and WW domain-binding protein 2(WBP2)in bladder cancer.Methods A total of 112 patients with bladder cancer admitted to Zhumadian Central Hospital from December 2017 to December 2019 were selected,and the cancer tissue specimens of bladder cancer patients(bladder cancer group)and the corresponding paracancerous tissue specimens(paracancerous group)were collected.The WWOX and WBP2 in bladder cancer tissues and adjacent tissues were measured by immunohistochemistry.The relationship between the WWOX,WBP2 and the clinicopathological characteristics of bladder cancer cases was explored byχ^(2)test.The factors affecting the prognosis of cases with bladder cancer were explored by multivariate Cox regression analysis.Results The positive rate of WBP2 in bladder cancer group was higher compared with adjacent group,and the positive rate of WWOX in bladder cancer group was lower compared with adjacent group(P<0.05).The positive expression rate of WBP2 and negative expression rate of WWOX in bladder cancer patients with low differentiation,tumor diameter≥1 cm,TNM stageⅢ-Ⅳ,lymph node metastasis and vascular space invasion were higher than those in bladder cancer patients with medium/high differentiation,tumor diameter<1 cm,TNM stageⅠ-Ⅱ,no lymph node metastasis and no vascular space invasion(P<0.05).The 3-year survival rate of patients with negative WWOX expression was higher than patients with positive WWOX expression(P<0.05).The 3-year survival rate of cases with positive WBP2 was lower compared with cases with negative WBP2(P<0.05).Ⅲ-Ⅳstage,lymph node metastasis,negative WWOX and positive WBP2 were risk factors for the prognosis of patients with bladder cancer(P<0.05).Conclusion WWOX is mainly negative expression in bladder cancer tissues and WBP2 is mainly positive expression.Both of them are closely related to the clinicopathological characteristics of bladder cancer patients,and are expected
