摘 要:目的:探讨氢气在非小细胞肺癌(non-small cell lung cancer,NSCLC)治疗中的作用及可能机制。方法:选取河南大学第一附属医院2018年1月至2020年12月首次确诊为NSCLC的患者60例,随机分为氢气组与对照组,两组均给予多西他赛+顺铂化疗一周期,期间氢气组每天吸入氢气2 h。观察两组患者的不良反应;干预前后检测两组患者肿瘤异常蛋白(tumor abnormal protein,TAP);干预后两组患者均取活检组织,HE染色观察肿瘤组织形态,免疫组化检测肿瘤组织中Ki-67、环氧化酶2(COX-2)、血管内皮生长因子(VEGF)的表达。结果:干预期间氢气组较对照组恶心、呕吐、骨髓抑制、脱发等不良反应减少,差异均有统计学意义(χ^(2)分别为4.59、5.08、5.93和4.44,P<0.05);干预后,氢气组较对照组TAP降低明显,差异有统计学意义(t=16.14,P<0.001),氢气组较对照组细胞异质性降低,Ki-67、COX-2、VEGF蛋白表达减少。结论:氢气可减少化疗引起的不良反应,且对NSCLC具有辅助治疗作用,可能与抑制Ki-67、COX-2、VEGF的表达有关。Objective:To nvestigate the role and potential mechanisms of hydrogen in the treatment of non-small cell lung cancer(NSCLC).Methods:60 patients who were newly diagnosed with non-small cell lung cancer(NSCLC)were selected and randomly divided into a hydrogen group and a control group.Both groups received one cycle of chemotherapy with docetaxel and cisplatin,during which the hydrogen group inhaled hydrogen for 2 hours each day.Adverse reactions were observed in both groups.Before and after the intervention,tumor abnormal protein(TAP)levels were detected in both groups.After the intervention,biopsy tissues were collected from both groups for hematoxylin and eosin(HE)staining to observe tumor tissue morphology.Immunohistochemical analysis was performed to detect the expression of Ki-67,cyclooxygenase-2(COX-2),and vascular endothelial growth factor(VEGF)in the tumor tissues.Results:During the intervention period,the adverse reactions such as nausea,vomiting,bone marrow suppression and alopecia in the hydrogen group were significantly lower than those in the control group(χ^(2)=4.59,5.08,5.93 and 4.44,P<0.05);After the intervention,the TAP and the cell heterogeneity were significantly lower in the hydrogen group than those in the control group,and the expressions of Ki-67,COX-2 and VEGF proteins were decreased.Conclusion:Hydrogen can reduce the adverse reactions caused by chemotherapy,and has adjuvant therapeutic effect on NSCLC,which may be related to the inhibitions of Ki-67,COX-2,VEGF expressions.Conclusion:Hydrogen can reduce adverse reactions caused by chemotherapy and has an adjuvant therapeutic effect on NSCLC,which may be related to its inhibition of the expression of Ki-67,COX-2,and VEGF.
