摘  要:目的研究核因子C1(nuclear factor C1,NFIC1)对皮肤鳞状细胞癌(cutaneous squamous cell carcinoma,CSCC)细胞活力、增殖、侵袭迁移及AKT相关信号通路蛋白的影响。方法用免疫组织化学检测NFIC1在正常皮肤和CSCC中表达差异;RT-q PCR检测NFIC1 m RNA在CSCC组织和细胞系中的表达。将NFIC1的过表达质粒和阴性质粒分别转染入CSCC细胞A431中,用MTT法、Ed U染色法和集落形成实验检测细胞增殖能力,用伤口愈合实验和Transwell分析检测细胞侵袭和迁移能力,用Western blot分析p-AKT、细胞周期素D1(cyclin D1)、神经钙黏蛋白(N-cadherin,N-CAD)和上皮钙黏蛋白(E-cadherin,E-CAD)蛋白水平。构建CSCC细胞移植瘤模型,观察过表达NFIC1对CSCC移植瘤生长的影响。结果NFIC1在CSCC组织和细胞系中表达下调。过表达NFIC1的A431细胞的增殖活力、迁移和侵袭能力显著下降,p-AKT、cyclin D1和N-CAD的水平下调,E-CAD水平上调。结论NFIC1能抑制CSCC细胞增殖、迁移和侵袭,此作用可能与其抑制p-AKT、cyclin D1、N-CAD表达和促进E-CAD表达有关。Objective To investigate the effects of nuclear factor C1(NFIC1)on cell viability,proliferation,invasion,migration,and AKT-related signaling pathway proteins in cutaneous squamous cell carcinoma(CSCC).Methods Immunohistochemistry was used to assess the differential expression of NFIC1 in normal skin and CSCC.RT-qPCR was employed to measure the expression of NFIC1 mRNA in CSCC tissues and cell lines of CSCC.NFIC1 overexpression plasmid and negative control plasmid were transfected into CSCC cells(A431).MTT assay,EdU staining,and colony formation assay were used to assess cell proliferation.Wound healing assay and Transwell assay were used to evaluate cell invasion and migration.Western blotting was performed to analyze the levels of p-AKT,cyclin D1,N-cadherin(N-CAD),and E-cadherin(E-CAD)proteins.A CSCC cell xenograft model was established to observe the effect of NFIC1 overexpression on the growth of CSCC tumors.Results NFIC1 expression was downregulated in the tissues and cell lines of CSCC.The proliferation,migration,and invasion abilities of NFIC1-overexpressing A431 cells were significantly reduced,with downregulation of p-AKT,cyclin D1,and N-CAD levels,and upregulation of E-CAD levels.Conclusion NFIC1 inhibits proliferation,migration,and invasion of CSCC cells,and this effect may be related to its inhibition of p-AKT,cyclin D1,N-CAD expression,and promotion of E-CAD expression.
