摘  要:目的探讨在DNA倍体分析中,甲苯胺蓝代替硫堇进行DNA染色的可能性。方法选取细胞学标本和手术标本各3例,每例制作2张涂/切片,1张用盐酸水解,1张不用盐酸水解,甲苯胺蓝染色液染色,检测甲苯胺蓝与DNA结合的特异性;选取细胞学标本3例,每例制作2张涂片,分别用甲苯胺蓝和硫堇染色液染色,在染色液配制后的第1 d、第2d和第5 d做3批对比实验,对比两种染色液的染色效果,并且用全自动病理细胞分析仪检测玻片IOD值和ID值。结果甲苯胺蓝可以特异性与DNA结合;甲苯胺蓝染色液染色效果优于硫堇染色液,有效期更长。结论甲苯胺蓝染色液可以用于DNA倍体分析实验,染色玻片经细胞分析仪检测后各项指标符合要求。Objective To investigate the possibility of using toluidine blue as a substitute for thionin in DNA ploidy analysis.Methods Three cytological specimens and three surgical specimens were selected,with two(smear)slides prepared for each specimen.One slide was hydrolyzed with hydrochloric acid,while the other was not.The slides were stained with toluidine blue to examine the specificity of DNA binding with toluidine blue.Additionally,three cytological specimens were selected,and two smear slides were prepared for each specimen.One slide was stained with toluidine blue,and the other with thionin.A comparison experiment was conducted on the 1st,2nd,and 5th day after the staining solutions were prepared.The staining effects of the two solutions were compared,and the slides were analyzed using an automatic pathological cell analyzer to measure the IOD(Integrated Optical Density)and ID(Intensity Density)values.Results Toluidine blue specifically binds to DNA.The staining effect of toluidine blue was superior to that of thionin,with a longer effective period.Conclusion Toluidine blue staining solution can be used in DNA ploidy analysis experiments.The slides stained with toluidine blue met the required standards after being analyzed by the cell analyzer.
