摘  要:目的:探讨新疆药桑叶提取物对人脐静脉内皮细胞(HUVECs)的成血管作用,为其在牙周组织再生领域的潜在应用提供理论依据。方法:离体培养HUVECs,制备新疆药桑叶提取物,CCK-8法检测不同浓度新疆药桑叶提取物对HUVECs增殖活性的影响,筛选出促进HUVECs细胞术的适宜浓度范围,分为空白对照组、药物组(62.5、125和250μg/mL桑叶提取物溶液)。采用流式细胞检测各组细胞凋亡率和活性氧表达水平;划痕实验和Transwell实验检测细胞迁移能力;小管形成实验检测细胞体外成管能力;RT-PCR检测细胞VEGF、bFGF、CD31、CD34 mRNA表达水平;免疫荧光染色检测细胞VEGF表达。结果:在62.5、125和250μg/mL桑叶提取物浓度下对细胞活力显著增强,差异有统计学意义(P<0.001)。与空白组比较,各浓度桑叶提取物对细胞凋亡无明显影响(P>0.05),各药物组细胞活性氧含量减少,相对水平迁移率、垂直迁移率、体外小管形成总长度、VEGF、bFGF、CD31、CD34 mRNA和VEGF蛋白表达均显著增强,差异有统计学意义(P<0.05)。结论:新疆药桑叶提取物可以促进HUVECs的增殖、迁移及体外小管形成,上调血管形成相关基因表达,减少氧化应激反应,进而促进HUVECs的血管生成能力,对其促进牙周组织再生具有潜在作用。AIM:Exploring the angiogenic effect of Xinjiang medicinal mulberry leaf extract on human umbilical vein endothelial cells(HUVECs)and providing theoretical basis for its potential application in periodontal tissue regeneration.METHODS:HUVECs were cultured in vitro to prepare Xinjiang medicinal mulberry leaf extract.The CCK-8 method was used to detect the effect of different concentrations of Xinjiang medicinal mulberry leaf extract on the proliferation activity of HUVECs.The appropriate concentration range for promoting HUVECs cell viability was screened and divided into blank control group and drug group(62.5,125 and 250μg/mL mulberry leaf extract solutions).Flow cytometry was used to detect the apoptosis rate and reactive oxygen species expression levels of each group of cells,scratch assay and Transwell assay were used to detect cell migration ability,tubular formation assay was used to detect the in vitro tube forming ability of cells,RT-PCR was used to detect the mRNA expression levels of VEGF,bFGF,CD31 and CD34 in cells,and immunofluorescence staining was used to detect the expression of VEGF in cells.RESULTS:The cell viability was significantly enhanced at concentrations of 62.5,125 and 250μg/mL mulberry leaf extract,with statistically significant differences(P<0.001).Compared with the blank group,mulberry leaf extract at different concentrations had no significant effect on cell apoptosis(P<0.05).The reactive oxygen species content of cells in each drug group decreased,and the relative horizontal migration rate,vertical migration rate,total length of in vitro tubular formation,VEGF,bFGF,CD31,CD34 mRNA and VEGF protein expression were significantly increased,with statistical significance(P<0.05).CONCLUSION:The extract of Xinjiang medicinal mulberry leaves can promote the proliferation,migration,and in vitro tubular formation of HUVECs,upregulate the expression of vascular formation related genes,reduce oxidative stress response,and thus promote the angiogenesis ability of HUVECs,which has potential effect
