摘　　要：目的分析环状核糖核酸转化/转录域关联蛋白(circTRRAP)对结直肠癌细胞生物学行为的影响。方法利用慢病毒转染结直肠癌细胞,将细胞分为circTRRAP组与NC组。RT-qPCR检测两组结直肠癌细胞中circTRRAP的表达量,通过体外实验分析两组结直肠癌细胞的增殖力、迁移力、侵袭力及细胞凋亡的变化。通过体内成瘤实验分析裸鼠皮下成瘤的变化(使用随机数字表法将12只6周龄BALB/c裸鼠分为过表达组和正常组,将大约1×10^(7)个过表达circTRRAP或正常的HCT116细胞皮下注射到过表达组和正常组裸鼠右腋窝区)。结果与NC组比较,circTRRAP组结直肠癌细胞中的circTRRAP mRNA相对表达量升高(P<0.05)。与NC组比较,circTRRAP组结直肠癌细胞的增殖、迁移及侵袭能力上升,细胞的凋亡水平下降(P<0.05)。与正常组比较,过表达组肿瘤体积和重量均增加(P<0.05)。结论circTRRAP促进了结直肠癌细胞恶性表型的生物学行为,可能发挥促癌作用。Objective To analyze the effects of circular RNA transformation/transcription domain associated protein(circTRRAP)on the biological behavior of colorectal cancer cells.Methods Colorectal cancer cells were transfected with lentivirus and divided into circTRRAP group and NC group.The expression level of circTRRAP in two groups of colorectal cancer cells was detected by RT-qPCR.The changes of proliferation,migration,invasion,and apoptosis of two groups of colorectal cancer cells were analyzed by in vitro experiments.The changes of subcutaneous tumorigenesis in nude mice were analyzed by in vivo tumorigenesis experiments(twelve six-week-old BALB/c nude mice were divided into overexpression group and normal group by random number table method,and about 1×10^(7) overexpression circTRRAP or normal HCT116 cells were subcutaneously injected into the right axillary area of nude mice in the overexpression group and the normal group).Results Compared with the NC group,the relative expression of circTRRAP mRNA in colorectal cancer cells in the circTRRAP group was increased(P<0.05).Compared with NC group,the proliferation,migration,and invasion ability of colorectal cancer cells in circTRRAP group increased,and the apoptosis level of cells decreased(P<0.05).Compared with the normal group,the tumor volume and weight of the overexpression group were increased(P<0.05).Conclusion The circTRRAP promotes the biological behavior of malignant phenotype of colorectal cancer cells and may play a role in promoting cancer.
