摘  要:采用超声辅助酶法提取山豆根生物碱,以山豆根生物碱提取量为响应值,通过单因素实验和响应面法优化了山豆根生物碱的提取工艺,并通过DPPH自由基和羟基自由基(·OH)清除实验评价了其抗氧化活性。结果表明,在纤维素酶加量为0.04 g、酶解时间为93 min、酶解温度为66℃、超声功率为175 W、乙醇体积分数为40%、超声温度为45℃、超声时间为30 min的最佳提取工艺下,山豆根生物碱提取量为10.568 mg·g^(-1);山豆根生物碱对DPPH自由基和·OH具有较强的清除能力,当浓度为0.05 mg·mL^(-1)时,清除率分别达到79.85%和84.24%。We extracted alkaloids from Sophora tonkinensis by a ultrasonic-assisted enzymatic method,and optimized the extraction process of alkaloids from Sophora tonkinensis through single-factor experiments and response surface methodologies by using the extraction amount of alkaloids as a response value.Moreover,we evaluated the antioxidant activity of alkaloids from Sophora tonkinensis through DPPH free radicals and hydroxyl free radicals(·OH)scavenging experiments.The results show that the extraction amount of alkaloids from Sophora tonkinensis can reach 10.568 mg·g^(-1) under the optimal extraction conditions as follows:the cellulase dosage of 0.04 g,the enzymolysis time of 93 min,the enzymolysis temperature of 66℃,the ultrasonic power of 175 W,the ethanol volume fraction of 40%,the ultrasonic temperature of 45℃,and the ultrasonic time of 30 min.In addition,alkaloids from Sophora tonkinensis have strong scavenging ability to DPPH free radicals and hydroxyl free radicals.When the concentration of alkaloids from Sophora tonkinensis is 0.05 mg·mL^(-1),the scavenging rates of DPPH free radicals and hydroxyl free radicals can reach 79.85% and 84.24%,respectively.
