摘  要:以紫球藻为原料,采用超滤结合一步疏水层析技术制备高纯度的藻红蛋白,分析了其可见光光谱、荧光发射光谱特征和亚基组成,并评价了其抗氧化活性。结果表明,纯化藻红蛋白的光谱学纯度(A545/A280)达到4.79;其在可见光区的545 nm和565 nm处有2个吸收峰,在498 nm处有1个肩峰,最大荧光发射峰位于580 nm处;纯化藻红蛋白由α、β和γ等3个亚基组成;纯化藻红蛋白具有一定的抗氧化活性,当其质量浓度为1.0 mg·mL^(-1)时,对羟基自由基、DPPH自由基的清除率分别达到62.97%、73.12%。该藻红蛋白制备工艺绿色高效,为其商品化生产和应用奠定了基础。We prepared high-purity phycoerythrin from Porphyridium cruentum by ultrafiltration combined with one-step hydrophobic chromatography.Moreover,we analyzed its visible light spectrum,fluorescence emission spectrum,and subunit composition,and evaluated its antioxidant activity.The results show that the spectral purity(A545/A280)of the purified phycoerythrin reaches 4.79.There are two absorption peaks at 545 nm and 565 nm,one acromion at 498 nm,and the maximum fluorescence emission peak is located at 580 nm.The purified phycoerythrin consists of three subunitsα,β,andγ.When the mass concentration of phycoerythrin is 1.0 mg·mL^(-1),the scavenging rate of hydroxyl free radicals and DPPH free radicals reach 62.97%and 73.12%,respectively,indicating that the purified phycoerythrin has a certain antioxidant activity.The preparation process of phycoerythrin is green and efficient,which lays a foundation for its commercial production and application.
