摘 要:建立高效液相色谱法测定月柿提取物中没食子酸含量的方法,为其作为化妆品用植物原料质量控制提供科学依据。采用CAPCELL PAK C_(18)MG(4.6 mm×250 mm,5μm)色谱柱,流动相为0.2%磷酸-甲醇,梯度洗脱,流速为1.0 mL/min,二极管阵列检测器检测,检测波长为273 nm,柱温30℃,进样量10μL。该色谱条件下,没食子酸在0.14~184.89 mg/L范围内呈良好的线性关系,R^(2)=0.9993,高、中、低3个质量浓度的加标回收率为88.58%~91.24%,RSD为0.19%~0.97%,方法精密度和准确度良好,供试品稳定性良好。分析结果表明,该方法学验证结果良好,样品主成分与其他成分分离效果好,操作简便,可用于月柿提取物中没食子酸含量的测定及作为化妆品原料标准中功效成分的质量控制方法。A method for determining the content of gallic acid in persimmon extract by high-performance liquid chromatography(HPLC)was established,providing a scientific basis for its quality control as a plant raw material for cosmetics.The chromatography was performed on a CAPCELL PAK C_(18) MG(4.6 mm×250 mm,5μm)column with the mobile phase consisting of 0.2% phosphoric acid-methanol and gradient elution at the flow rate of 1.0 mL/min,the detection wavelength was 273 nm,the column temperature was 30℃,and the injection volume was 10μL.Under this chromatographic condition,gallic acid showed a good liner relationship in the concentration range of 0.14~184.89 mg/L,R^(2)=0.9993.The recovery of high,medium and low concentration was from 88.58% to 91.24%,RSD was 0.19%~0.97%.The method has good precision and accuracy,and the stability of the test sample is good.The analysis results show that the method has good methodological validation results,good separation effect from other components,simple operation,can be used for the determination of gallic acid content in persimmon extract and as a quality control method for effective ingredients in the standard of cosmetic raw materials.
