摘  要:本研究基于加权基因共表达网络分析(weighted correlation network analysis,WGCNA)联合基因表达综合数据库(gene expression omnibus,GEO)鉴定共表达基因,经癌症基因组图谱(the cancer genome atlas,TCGA)验证共表达基因在宫颈癌中的表达并确定关键基因。结果显示,突触蛋白3(synaptogyrin 3,SYNGR3)为宫颈癌的关键基因,受试者工作特征曲线(receiver operating characteristic curve,ROC)的曲线下的面积显示SYNGR3预测宫颈癌的准确性为99.1%。Cox回归结果显示,高表达SYNGR3的宫颈癌患者生存周期延长。免疫浸润显示,高表达SYNGR3和Th1细胞的免疫浸润相关。人类蛋白质图谱数据库中免疫组织化学染色结果显示,SYNGR3在宫颈腺癌中表达。蛋白免疫印迹和实时定量荧光PCR实验结果显示,SYNGR3在人宫颈癌细胞C-33a、人宫颈癌细胞HeLa及人宫颈鳞状细胞癌细胞Siha中高表达。本实验结果显示,SYNGR3可以作为为宫颈癌潜在的生物标志物且与宫颈癌患者的总生存期相关。This research bases on weighted correlation network analysis(WGCNA)in combination with the gene expression omnibus(GEO)to identify co-expressed genes.The expression of the co-expressed genes in cervical cancer was then verified through the cancer genome atlas(TCGA),and the key genes were determined.The results indicated that synaptogyrin 3(SYNGR3)is a crucial gene in cervical cancer.The area under the receiver operating characteristic curve(ROC)demonstrated that the accuracy of SYNGR3 in predicting cervical cancer was 99.1%.The results of Cox regression demonstrated that cervical cancer patients with high expression of SYNGR3 had a prolonged survival period.Immunoinfiltration analysis exhibited that high SYNGR3 expression was correlated with the immunoinfiltration of Th1 cells.The immunohistochemical staining results in the Human Protein Atlas database showed that SYNGR3 was expressed in cervical adenocarcinoma.The results of Western blot and real-time quantitative fluorescence PCR experiments indicated that SYNGR3 was highly expressed in human cervical cancer cell line C-33a,human cervical cancer cell line HeLa,and human cervical squamous cell carcinoma cell line Siha.SYNGR3 is a potential biomarker for cervical cancer and is associated with the overall survival of cervical cancer patients.
