摘  要:目的:探讨不同移植方式下人脱落乳牙牙髓干细胞(SHED)对Wistar-Kyoto(WKY)抑郁模型大鼠的治疗作用。方法:将30只雄性WKY大鼠随机分为模型组、脑室注射组、经鼻递送组,每组10只;另有10只雄性Wistar大鼠作为对照组。WKY大鼠适应性饲养1周后分别给予脑室内注射SHED细胞和鼻内递送SHED细胞,移植3周后通过蔗糖偏好实验、Y迷宫实验、强迫游泳实验及旷场实验评估不同移植方式下SHED治疗的效果;酶联免疫吸附试验(ELISA)检测神经营养因子BDNF和炎症相关因子IL-1β、IL-6、IL-10及TNF-α等的表达水平。结果:行为学实验观察到SHED移植治疗可以减轻WKY抑郁模型大鼠的抑郁样行为并提高大鼠短期记忆力,脑室注射组的治疗作用高于经鼻递送组;ELISA试验检测到SHED移植治疗后大鼠海马区和血清中神经营养因子BDNF和抗炎因子IL-10表达水平升高,而促炎因子IL-1β、IL-6及TNF-α等的表达水平降低,脑室注射组的治疗作用高于经鼻递送组。结论:SHED移植治疗具有抗抑郁作用,可能是通过神经营养保护和免疫调节抗炎的方式实现,脑室内移植治疗的作用优于经鼻递送。Objective:To investigate the therapeutic effects of stem cells from human exfoliated deciduous teeth(SHED)transplanted by different methods on the Wistar-Kyoto(WKY)depression model rats.Methods:30 male WKY rats were randomly divided into three groups:the model group,intraventricular injection group,and intranasal delivery group,with 10 rats in each group.10 male Wistar rats served as the control group.The WKY rats was acclimated for 1 week before receiving either intraventricular injection or intranasal delivery of SHED cells.3 weeks after transplantation,the therapeutic effects of SHED transplantation were evaluated using sucrose preference test,Y-maze experiment,forced swimming test,and open field test.Additionally,enzyme-linked immunosorbent assay(ELISA)was used to detect the expression levels of neurotrophic factor BDNF and inflammation-related factors IL-1β,IL-6,IL-10,and TNF-α.Results:Behavioral experiments demonstrated that SHED transplantation alleviated depressive-like behaviors and improved short-term memory in WKY depression model rats.The therapeutic effect of intraventricular injection was superior to intranasal delivery.ELISA experiments revealed increased expression of neurotrophic factor BDNF and anti-inflammatory factor IL-10 in the hippocampus and serum following SHED transplantation;meanwhile pro-inflammatory factors IL-1β,IL-6,and TNF-αshowed decreased expression.Again intraventricular injection exhibited a more pronounced therapeutic effect compared to intranasal delivery.Conclusion:SHED transplantation therapy has an antidepressant effect possibly achieved through neural nutritional protection and immune regulation against inflammation pathways,and intraventricular transplantation appears more effective than intranasal delivery.
