摘 要:目的:探讨宫颈鳞癌(CSCC)中tRF-Phe48的表达及其临床病理意义。方法:采用qRT-PCR法检测CSCC中tRF-Phe48的表达水平。采用RNA原位分子杂交(RISH)检测79对CSCC组织和相邻健康组织中tRF-Phe48的阳性表达,并研究其与临床特征的Spearman相关性。采用CCK-8实验评估细胞增殖。生物信息学预测tRF-Phe48的靶基因并通过qRT-PCR验证。结果:qRT-PCR检测显示,tRF-Phe48在CSCC中表达水平明显高于癌旁正常组织(P<0.05)。RISH结果显示,在CSCC细胞中tRF-Phe48染成棕黄色,为阳性表达。tRF-Phe48阳性表达与肿瘤大小密切相关(P<0.05),而与患者年龄、HPV感染、分化、转移及TNM分期无关(P均>0.05)。CCK-8实验显示过表达tRF-Phe48增强CSCC细胞的增殖活性,而降低tRF-Phe48表达则抑制细胞增殖活性(P均<0.05)。生物信息学预测FKBP9、NETO1和PRKDC是tRF-Phe48的下游靶基因,qRT-PCR验证发现无论过表达tRF-Phe48还是降低tRF-Phe48表达,FKBP9、NETO1和PRKDC的表达水平变化不大。结论:在CSCC中tRF-Phe48表达水平上调,且与肿瘤大小密切相关。tRF-Phe48是一个促进细胞增殖的癌基因,有望成为CSCC增殖的生物标志物。Objective:The expression of tRF-Phe48 in cervical squamous cell carcinoma(CSCC)and its clinicopathological significance were discussed.Methods:The expression of tRF-Phe48 in CSCC was detected by qRT-PCR.RNA in situ hybridization(RISH)was used to detect the positive expression of tRF-Phe48 in 79 CSCC tissues,and to study its Spearman correlation with clinical characteristics.Cell proliferation was evaluated by CCK-8.The target gene of tRF-Phe48 was predicted by bioinformatics and verified by qRT-PCR.Results:qRT-PCR showed that the expression level of tRF-Phe48 in CSCC was significantly higher than that in adjacent healthy tissues(P<0.05).RISH results showed that tRF-Phe48 was positively expressed in CSCC cells with brownish-yellow color.The positive expression of tRF-Phe48 was closely correlated with tumor size(P<0.05),but not with age,HPV infection,differentiation,metastasis and TNM stage(all P>0.05).CCK-8 assay showed that overexpression of tRF-Phe48 enhanced the proliferation activity of CSCC cells,while decreased expression of tRF-Phe48 inhibited the proliferation activity of CSCC cells(all P<0.05).Bioinformatics predicted that FKBP9,NETO1 and PRKDC were the downstream target genes of tRF-Phe48,and qRT-PCR verified that the expression levels of FKBP9,NETO1 and PRKDC were not significantly changed whether tRF-Phe48 was overexpressed or reduced.Conclusion:The expression level of tRF-Phe48 is up-regulated in CSCC and is closely related to tumor size.tRF-Phe48 is an oncogene that promotes cell proliferation and is expected to be a biomarker for CSCC proliferation.
