摘  要:试验旨在研究槟榔提取物(ANE)及其四种主要生物碱(槟榔碱、槟榔次碱、去甲槟榔碱和去甲槟榔次碱)对洋刀豆脲酶(JBU)的抑制效果,选用乙酰氧肟酸(AHA)作为对照,并深入探究其抑制作用位点。结果显示,去甲槟榔碱(Guv)对JBU抑制效果最显著,IC_(50)值为31.06 mg/L。酶动力学评价结果显示,ANE和Guv均为JBU的混合型抑制剂。硫醇试剂存在时,ANE和Guv对JBU的抑制效果显著降低(P<0.05);Ni^(2+)竞争性抑制剂存在时,Guv对JBU的抑制效果显著提高(P<0.05)。分子对接结果显示,Guv与氨基酸残基Asp 494、Ala 440、His 519、His 593、Arg 609、Gly 550、His 492、Ala 636、Asp 633和His 409形成相互作用。研究表明,Guv能够有效减少JBU对尿素的分解,槟榔具备新型脲酶抑制剂功能产品开发的潜能。The experiment aims to investigate the inhibitory effects of Arecacatechu extract(ANE)and its four main alkaloids(arecoline,arecaidine,guvacine,and guvacoline)on jack bean urease(JBU),using acetoxyhydroxamic acid(AHA)as a control,and to further explore the sites of their inhibitory action.The results showed that guvacoline(Guv)exhibited the most significant JBU inhibition effect with an IC_(50) value of 31.06 mg/L.Urease kinetics experiments showed that both ANE and Guv are hybrid inhibitors of JBU.The inhibitory effects of ANE and Guv on JBU were significantly reduced in the presence of thiol reagents(P<0.05),and the inhibitory effects of Guv on JBU were significantly elevated in the presence of Ni^(2+)competitive inhibitors(P<0.05).Molecular docking results showed that Guv formed interactions with amino acid residues Asp 494,Ala 440,His 519,His 593,Arg 609,Gly 550,His 492,Ala 636,Asp 633,and His 409.The study indicates that Guv can effectively reduce the catabolism of urea by JBU,and Arecacatechupossesses the potential for novel urease inhibitor functional product development.
