摘 要:目的 探索不同溶剂对丹参酮ⅡA光降解速率的影响及降解动力学特征。方法 采用紫外-可见分光光度法(UV-VIS)测定丹参酮ⅡA的吸光值,研究丹参酮ⅡA在不同溶剂中的光降解情况。结果 各溶剂中丹参酮ⅡA在避光环境下的降解率仅为1%左右,在8000 LUX照度恒定情况下,丹参酮ⅡA的光降解均符合一级动力学反应,且在不同溶剂中丹参酮ⅡA的降解速率差别较大,半衰期分别为甲醇9.1 h、乙腈3.4 h、乙醇2.4 h、丙酮0.7 h、乙酸乙酯0.2 h、乙醚0.1 h。结论 丹参酮ⅡA在不同溶剂中均发生了光降解,且降解速率会随着溶剂极性的增大而降低,推测丹参酮ⅡA经光照后自身裂解形成激发态,在极性溶剂介质中发生反应形成了稳定的自由基产物,从而降低了丹参酮ⅡA的光降解速率。提示在有丹参酮ⅡA存在的生产过程中应尽量避光,采用醇类溶剂能增加其稳定性,避免有效成分损失。Objective To explore the effects of different solvents on the photodegradation rate of tanshinone IIA and the characteristics of degradation kinetics.Methods The absorbance values of tanshinone IIA were determined by ultraviolet-visible spectrophotometry(UV-VIS)to study the photodegradation of tanshinone IIA in different solvents.Results The degradation of tanshinone IIA in each solvent was only about 1%in the light-avoidance environment,and the photodegradation of tanshinone IIA was in accordance with the first-order kinetic reaction under the constant illumination of 8000 LUX,and the degradation rate of tanshinone IIA varied greatly among different solvents,with the half-lives of 9.1h in methanol,3.4 h in acetonitrile,2.4h in ethanol,0.7 h in acetone,0.2 h in ethyl acetate,and 0.2 h in ether,respectively,0.1 h for ethyl ether.Conclusion Tan-shinone IIA is photodegraded in different solvents,and the degradation rate would be reduced with the increase of solvent polari-ty,which is presumed to be the formation of excited state by the cleavage of tanshinone IIA by light itself,and the formation of stable radical products by the reaction in the polar solvent medium,which reduces the photodegradation rate of tanshinone IIA.It is suggested that light should be avoided as much as possible during the production process in the presence of tanshinone IA,and the use of alcoholic solvents can increase its stability and avoid the loss of active ingredients.
