摘 要:为了探究鸭绿江入海口湿地中土壤细菌的群落多样性、结构及其组装机制,利用高通量测序技术对滩涂盐土、碱蓬草盐土、沼泽土、水稻土、草甸土和棕壤等6种典型土壤的细菌群落进行16S rRNA基因扩增子测序,并基于系统发育二进制的零模型分析(model analysis)解析细菌群落组装机制。结果表明,变形菌门(Proteobacteria)平均相对丰度最高(38.5%),其次为酸杆菌门(Acidobacteria,18.2%)和拟杆菌门(Bacteroidetes,9.7%)。6种土壤类型细菌群落的α多样性和β多样性整体差异显著(P<0.05)。主要菌门和菌属与环境因子的冗余分析(RDA)结果表明,土壤有机碳(SOC)、全氮(TN)、电导率(EC)、全硫(TS)和pH是主要影响因子。基于零模型的群落组装分析表明,鸭绿江口湿地土壤细菌群落组装以随机性为主导,其中扩散限制贡献最大,在群落组装中占40.52%。研究结果可为鸭绿江口湿地生态管理提供参考。In order to explore the community diversity,structure and assembly mechanism of soil bacteria in the Yalu River estuary wetland,high-throughput sequencing technology was used to sequence 16SrRNA gene of bacterial communities forsix main representative soil types including tidal flat saline soil(TFSS),Suaeda salsa saline soil(SSSS),bog soil(BS),paddy soil(PS),meadow soil(MS)and brown forest soil(BFS)and inferring community assembly mechanisms by phylogenetic bin-based null model analysis(iCAMP)was used to elucidate the bacteria community assembly mechanism.The results showed that Proteobacteria(38.5%of total sequences)was the most abundant phylum,the other phyla with high abundance were Acidobacteria(18.2%)and Bacteroidetes(9.7%).The overall differences inαdiversity andβdiversity among the bacterial communities of the six major soil types were significant(P<0.05).The results of redundancy analysis of major bacteria and environmental factors showed that soil organic carbon(SOC),total nitrogen(TN),conductivity(EC),total sulfur(TS)and pH were the main influencing factors.The null model-based community assembly analysis showed that the soil bacterial community assembly in the Yalu River estuary wetland was dominated by randomness processes,accounting for 79.09%of the community assembly,of which dispersal limitation contributed the most,accounting for 40.52%.The results revealed the differences in the diversity and structure of soil bacterial community,the relationship between environmental factors and bacterial community structure and the mechanism of bacterial community assembly,which could provide an important reference for the ecological management of Yalu River Estuary wetland.
