摘 要:肉碱棕榈酰转移酶(CPT)是线粒体膜上调控细胞脂肪酸氧化的酶,可以将脂酰辅酶A运输到线粒体内,以便进行β氧化。CPT有两个亚型,分别为CPT1和CPT2。CPT1是调节肉碱跨线粒体内膜转运的关键酶,位于线粒体外膜上,CPT1缺乏可导致脂肪酸氧化率降低,引发异常脂肪酸氧化和肿瘤能量代谢重编程。CPT2位于细胞线粒体内膜上,是长链脂肪酸转运至线粒体进行β氧化的限速酶,是一种广泛分布的蛋白质,参与形成酰基辅酶A,CPT2缺乏通常会诱发一些脂质代谢疾病。近年来的研究指出,CPT2在肝癌、乳腺癌、卵巢癌、结肠癌的进展中起着重要的作用,相关研究及报道也众多。CPT2在肺腺癌中的表达及作用尚未研究清楚。所以,探究CPT2与肺腺癌之间潜在的病理机制,探索CPT2在肺腺癌中的表达及作用机制,为肺腺癌的临床诊疗提供新的思路。Carnitine palmitoyl transferase(CPT)is an enzyme on the mitochondrial membrane that regulates cellular fatty acid oxidation.It can transport fatty acyl coenzyme A to the mitochondria forβ-oxidation.Among these two subtypes,CPT1 plays an important role because it is a key enzyme that regulates carnitine transport across the inner membrane of mitochondria and is located on the outer membrane of mitochondria.CPT1 deficiency can lead to the decrease of fatty acidβoxidation rate,abnormal fatty acid oxidation and tumor energy metabolism reprogramming.CPT2,located in the mitochondrial inner membrane of the CPT family,is a rate-limiting enzyme that transports long-chain fatty acids to mitochondria forβ-oxidation.It is a widely distributed protein that participates in the formation of acyl-CoA,and its deficiency usually leads to some lipid metabolic diseases.In recent years,some studies have pointed out that CPT2 plays an important role in the progression of liver cancer,breast cancer,ovarian cancer and colon cancer.However,there are few reports about CPT2 and CPT1 in cancer,and its expression and role in lung adenocarcinoma have not been clearly studied.Therefore,exploring the potential pathological mechanism between it and lung adenocarcinoma and exploring the expression and mechanism of CPT2 in lung adenocarcinoma will provide new ideas for clinical diagnosis and treatment of lung adenocarcinoma.
