摘 要:目的 研究不同克隆号的Ki67抗体在不同肿瘤组织上的表达差异。方法 收集3种肿瘤的45例石蜡组织标本,每种肿瘤组织15例,制备多组织芯片。运用免疫组化技术,选取三种不同克隆号的Ki67:MIB-1、SP6、MXR002,其他条件保持一致,进行免疫组化实验。正常组织对照包括扁桃体、阑尾和脂肪组织。结果Ki67(MIB-1)在乳腺肿瘤、肺癌和子宫颈肿瘤的增殖指数分别为66.7%(10/15)、80%(12/15)和86.7%(13/15),染色强度为弱到中等染色;Ki67(SP6)在乳腺肿瘤、肺癌和宫颈肿瘤的增殖指数为60%(9/15)、73.3%(11/15)和73.3%(11/15),染色程度为轻至中等;在乳腺癌、肺癌和宫颈癌中,Ki67(MXR002)的增殖指数均达到100%(15/15),染色强度为强染色,高于Ki67(MIB-1)和Ki67(SP6)(P<0.05)。在常规扁桃体组织和阑尾组织中,Ki67(MXR002)的染色程度及增殖指数也高于Ki67(MIB-1)和Ki67(SP6)(P<0.05)。在脂肪组织样本中,Ki67(MIB-1)抗体出现非特异性染色,而Ki67(MXR002)和Ki67(SP6)抗体则未表现出此类现象。结论 Ki67(MIB-1)和Ki67(SP6)的敏感性均较低,其中Ki67(MIB-1)在脂肪细胞上呈现非特异性染色。因此,推荐采用Ki67(MXR002)进行免疫组化染色。Objective To investigate the expression differences of Ki67 antibodies with different clone numbers in various tumor tissues.Methods A total of 45 paraffin-embedded tissue specimens from three types of tumors were collected,with 15 specimens from each tumor type.A multi-tissue array was prepared.Immunohistochemical staining was performed using three different clones of Ki67:MIB-1,SP6,and MXR002,with all other conditions held constant.Normal tissue controls included tonsil,appendix,and adipose tissue.Results The proliferation indices of Ki67(MIB-1)in breast cancer,lung cancer,and cervical cancer were 66.7%(10/15),80%(12/15),and 86.7%(13/15),respectively,with weak to moderate staining intensity.The proliferation indices of Ki67(SP6)in breast cancer,lung cancer,and cervical cancer were 60%(9/15),73.3%(11/15),and 73.3%(11/15),respectively,with light to moderate staining intensity.In breast cancer,lung cancer,and cervical cancer,Ki67(MXR002)showed a proliferation index of 100%(15/15),with strong staining intensity,which was significantly higher than that of Ki67(MIB-1)and Ki67(SP6)(P<0.05).In normal tonsil and appendix tissues,Ki67(MXR002)also exhibited higher staining intensity and proliferation index compared to Ki67(MIB-1)and Ki67(SP6)(P<0.05).In adipose tissue samples,Ki67(MIB-1)antibody exhibited nonspecific staining,while Ki67(MXR002)and Ki67(SP6)antibodies did not show this phenomenon.Conclusion Ki67(MIB-1)and Ki67(SP6)have lower sensitivity,with Ki67(MIB-1)showing nonspecific staining in adipocytes.Therefore,Ki67(MXR002)is recommended for immunohistochemical staining.
