摘  要:目的:分析乳腺癌(breast cancer, BRCA)中RNA聚合酶ⅡJ亚基(RNA polymeraseⅡsubunit J,POLR2J)的表达和临床意义,并探究其在BRCA中发挥的生物学作用。方法:在多个数据库中分析POLR2J在细胞和组织层面的表达。使用免疫印迹分析POLR2J在BRCA细胞系中的表达。在多个数据库中分析POLR2J对BRCA预后和临床特征的影响。使用cibersort函数分析POLR2J对BRCA免疫浸润的影响。使用maftools包分析POLR2J在BRCA中的突变景观。使用limma包获取POLR2J差异表达基因,使用cor.test函数鉴定POLR2J相关基因,对交集基因进行蛋白质-蛋白质互作网络的绘制和功能富集分析。然后选取2021年1月至2023年12月在本院手术治疗的BRCA患者71例,收集患者病理组织标本,通过实时荧光定量PCR(qRT-PCR)以及免疫组织化学染色法检测POLR2J的癌组织和癌旁组织表达水平及与患者临床病理参数关系。结果:POLR2J在BRCA中高表达,且与BRCA的不良预后正相关。POLR2J的高表达预示着BRCA中雌激素受体、孕激素受体和HER2的阴性表达。POLR2J高表达与POLR2J低表达的BRCA具有明显不同的免疫微环境。通路富集分析显示,POLR2J与BRCA的氧化磷酸化,线粒体基因表达和电子传递链等功能相关。POLR2J高表达的BRCA具有更高的TP53突变率。POLR2J的高表达预示着更高的TNM分期和Grade分级。与癌旁组织相比,BRCA组织中POLR2J mRNA和蛋白水平显著升高。结论:POLR2J或许是预测BRCA预后的生物标志物,并影响BRCA的氧化磷酸化、免疫微环境和基因突变。Objective:To analyze the expression and clinical significance of RNA polymerase II subunit J(POLR2J)in breast cancer(BRCA),and investigate its biological roles in BRCA.Methods:POLR2J expression was analyzed at cellular and tissue levels in multiple databases.POLR2J expression in BRCA cell lines was analyzed using immunoblotting.The effect of POLR2J on prognosis and clinical features of BRCA was analyzed in multiple databases.We analyzed the effect of POLR2J on immune infiltration in BRCA using the cibersort function,and the mutational landscape of POLR2J in BRCA using the maftools package.Differentially expressed POLR2J genes were obtained using the limma package,POLR2J-related genes were identified using the cor.test function,and the intersecting genes were drawn and analyzed by protein-protein interaction networks and functional enrichment analysis.Then,71 BRCA patients who underwent surgical treatment in our hospital from January 2021 to December 2023 were selected,and the pathological tissue specimens of the patients were collected.The expression levels of POLR2J in cancerous and paracancerous tissues were detected by real-time fluorescence quantitative PCR and immunohistochemistry staining,and their relationships to the clinicopathological parameters of the patients were detected.Results:POLR2J was highly expressed in BRCA and was positively correlated to the poor prognosis of BRCA.The high expression of POLR2J predicted the negative expressions of estrogen receptor,progesterone receptor and human epidermal growth factor receptor 2 in BRCA.BRCA with high expression of POLR2J had a distinctly different immune microenvironment from that of BRCA with low expression of POLR2J.Pathway enrichment analysis showed that POLR2J was associated with oxidative phosphorylation,mitochondrial gene expression,and electron transport chain in BRCA.BRCA with high POLR2J expression had a higher TP53 mutation rate.High POLR2J expression was predictive of higher TNM stages and grades.POLR2J mRNA and protein levels were significant
