摘 要:耐药性是肿瘤治疗的重大挑战。肿瘤微环境中的癌细胞和多种基质细胞产生的外泌体,通过诸如药物外流、传递耐药相关分子和核酸以及调控抗肿瘤免疫反应等方式,参与了实体瘤靶向治疗耐药性的形成。同时,外泌体凭借其高效的负载能力和良好的毒性耐受性,展现出对抗肿瘤耐药性的潜在应用价值。本综述旨在总结并探讨外泌体在实体瘤靶向治疗中介导耐药性的机制以及它们在应对肿瘤耐药性中所扮演的新兴角色。Drug resistance is a major challenge in cancer treatment.Exosomes released by cancer cells within the tumor microenvironment and various stromal cells participate in the development of solid tumors resistant to targeted therapy through mechanisms such as drug efflux,transmission of drug-resistant molecules and nucleic acids,and modulation of anti-tumor immune responses.Moreover,due to their efficient cargo-loading capacity and excellent toxicity tolerance,exosomes exhibit potential applicability in overcoming cancer drug resistance.This review aims to summarize and discuss the mechanisms through which exosomes mediate resistance in solid tumor targeted therapy and their emerging roles in addressing cancer resistance.
