摘 要:为探究大黄鱼(Larimichthys crocea)抗流性状形成的生理机制,借助自制跑道式水槽装置,在水平流速1.0 m·s^(-1)条件下,筛选得到强抗流组(HM组,顶流时间>30 min)和弱抗流组(SM组,顶流时间<5 min),并比较分析了两组鱼抗氧化能力、肌肉超微结构及肌纤维相关基因表达的特征。结果显示:1)HM组肝脏总超氧化歧化酶(T-SOD)活性显著高于SM组(P<0.05),HM组鳃总抗氧化酶(T-AOC)和T-SOD活性显著高于SM组(P<0.05);2)使用透射电镜对肌肉观察发现,HM组肌原纤维直径、肌原纤维横截面积显著大于SM组(P<0.05),但是肌节缩短(P<0.05),且SM组肌原纤维较为松散;3)肌纤维相关基因qPCR结果显示,HM组igf-1、col7a1、myosin-7、mmp2、ppar基因的相对表达水平极显著高于SM组(P<0.01),而SM组ubiquitin基因的表达量显著高于HM组(P<0.05)。结果表明,抗流能力强的大黄鱼具有较发达的肌纤维结构和较强的抗氧化能力。研究结果为揭示大黄鱼抗流性状的生理机制奠定了基础,并可为进一步开展大黄鱼抗流性状的遗传选育提供数据支持。Large yellow croaker(Larimichthys crocea)represents the seawater fish with the largest aquaculture scale and highest yield in China.In recent years,deep-sea mariculture of L.crocea has emerged as a crucial strategy to alleviate pressure on the ecological environment of offshore waters and foster green,high-quality development within the industry.L.crocea samples collected from the Shacheng Bay aquaculture area in Fuding,Fujian Province were selected and screened as HM group(time against the flow>30 min)and SM group(time against the flow<5 min)by self-made raceway system with the flow rate of 1.0 m·s^(-1).The change trend of tissue antioxidant capacity,muscle ultrastructure and the expression of muscle fiber-related genes between HM group and SM group were compared and analyzed.The results showed that:1)The activity of T-SOD in liver of HM group L.crocea was significantly higher than that of SM group(P<0.05);activities of TAOC and T-SOD in muscle were not significantly different between two groups(P>0.05);activities of T-AOC and T-SOD in gill of HM group was significantly higher than that of SM group(P<0.05).2)The results of transmission electron microscopy showed that the basic structure of sarcomere in HM group was clearly visible,the sarcoplasmic reticulum was complete,and the myofibril was arranged evenly and neatly,while the myofibril in SM group was loose.The myofibril diameter and myofibril area in HM group were significantly higher than those in SM group(P<0.05),while the sarcomere length in HM group was significantly lower than that in SM group(P<0.05).3)The qPCR of muscle tissue results showed that the relative expression of igf-1,col7a1,myosin-7,mmp2,ppar of HM group were significantly higher than that in SM group(P<0.01),while the expression of ubiquitin was the opposite.The strength of the anti-flowing ability in L.crocea was significantly related to its physiological and biochemical state.Individuals with stronger anti-flowing ability exhibited enhanced antioxidant capacity,larger muscle fiber diame
