摘  要:2′脱氧腺苷是生物体内DNA的结构片段,通过对其结构中的碱基和脱氧核糖的修饰而形成的化学中间体,是制备多种抗病毒、抗肿瘤和抗艾滋病药物的重要原料。笔者以凝结芽孢杆菌全细胞催化合成2′脱氧腺苷,并对细胞添加量、反应温度、n(脱氧胸苷)∶n(腺嘌呤)、底物质量浓度、磷酸缓冲盐浓度和pH等参数进行了单因素实验和响应面优化,优化后底物转化率最高达85.9%,2′脱氧腺苷质量浓度为44.59 g/L,脱氧胸苷转化率提高了51.7%,产物的生成效果显著。温度、pH、细胞添加量和磷酸缓冲盐浓度对转化率的影响由大到小,验证实验结果与响应面分析结果一致,该研究可为2′脱氧腺苷的放大生产提供参考。2′deoxyadenosine is a structural fragment of DNA in vivo.It is a chemical intermediate formed by modifying the bases and deoxyribose in its structure,and is also an important raw material for preparing various antiviral,anticancer and anti-AIDS drugs.The author used Bacillus coagulans as a whole cell catalyst to synthesize 2′deoxyadenosine,and conducted single factor experiments and response surface optimization on parameters such as cell addition amount,reaction temperature,n(dT)∶n(A),substrate mass concentration,phosphate buffer concentration,and reaction pH.After optimization,the substrate conversion rate reached 85.9%,the mass concentration of 2′deoxyadenosine was 44.59 g/L,and the deoxythymidine conversion rate increased by 51.7%,the generation effect of the product is significant.The effects of temperature,pH,cell addition amount,and phosphate buffer concentration on conversion rate vary from large to small.The validation experiment results are consistent with the response surface analysis results.This study provides a reference for the scaled-up production of 2′deoxyadenosine.
