摘 要:目的探究不同宫颈组织中血管运动素样蛋白1(angiomotin-like protein 1,AMOTL1)与Hippo信号通路下游相关分子Yes相关蛋白(yes-associated protein,YAP)以及TAZ的表达及临床意义。方法选取2017年1月至2018年7月于徐州医科大学附属医院行手术治疗并经病理确诊的宫颈癌患者102例、宫颈鳞状上皮高级别病变(how-grade squamous intraepithelial lesion,HSIL)患者50例、低级别病变(low-grade squamous intraepithelial lesion,LSIL)患者45例和慢性宫颈炎患者50例。采用免疫组织化学方法检测各组宫颈组织中AMOTL1与Hippo通路下游相关蛋白YAP、TAZ的表达情况,并分析其与宫颈癌患者临床病理特征间的关系;计数资料组间率比较采用χ^(2)检验;采取Spearman相关分析法分析AMOTL1、YAP、TAZ间关系;采用Kaplan-Meier曲线进行生存分析;采用多因素Cox回归分析影响宫颈癌患者预后的因素。结果免疫组化结果显示,AMOTL1在慢性宫颈炎、LSIL、HSIL以及宫颈癌组织中阳性率依次为12.00%(6/50)、17.78%(8/45)、28.00%(14/50)、76.47%(78/102),宫颈癌组织中AMOTL1阳性表达高于其他组别(χ^(2)=82.54,P<0.001);YAP在慢性宫颈炎、LSIL、HSIL以及宫颈癌组织阳性率依次为10.00%(5/50)、15.56%(7/45)、30.00%(15/50)、63.73%(65/102),宫颈癌组织中YAP阳性表达高于其他组别(χ^(2)=56.66,P<0.001);TAZ在慢性宫颈炎、LSIL、HSIL以及宫颈癌组织中阳性率依次为8.00%(4/50)、17.78%(8/45)、30.00%(15/50)、71.57%(73/102),宫颈癌组织中TAZ阳性表达高于其他组别(χ^(2)=74.71,P<0.001)。在宫颈癌组织中,AMOTL1阳性表达与国际妇产科协会(Federation International of Gynecology and Obstetrics,FIGO)分期(χ^(2)=16.28,P=0.001)、组织分化程度(χ^(2)=30.16,P<0.001)及有无淋巴结转移有关(χ^(2)=5.81,P=0.016),YAP阳性表达与FIGO分期(χ^(2)=15.99,P<0.001)、分化程度(χ^(2)=25.06,P<0.001)、肿瘤直径(χ^(2)=13.63,P=0.003)及有无淋巴结转移有关(χ^(2)=5.78,P=0.016),TAZ阳性表Objective To investigate the expression and clinical significance of angiomotin-like protein 1(AMOTL1)and downstream related molecules of Hippo signaling pathway yes-associated protein(YAP)and TAZ in different cervical tissues.MethodsFrom January 2017 and July 2018,102 cervical cancer tissues,50 cervical squamous epithelial high-grade lesions(HSIL)tissues,45 low-grade lesions(LSIL)tissues and 50 chronic cervicitis tissues,all of which underwent surgical treatment and pathological diagnosis at Xuzhou Medical University Affiliated Hospital from January 2017 to July 2018,were selected.The expression of AMOTL1 and Hippo pathway downstream related proteins YAP and TAZ were evaluated by immunohistochemistry and their relationship with the clinicopathological parameters of cervical cancer were examined in the cervical tissues of each group.The comparison of inter group rates in count data was conducted using the chi square test.Adopting Spearman correlation analysis method to analyze the relationship between AMOTL1,YAP,and TAZ.Single factor survival analysis was performed using Kaplan Meier curves.Kaplan Meier method and multivariate Cox regression analysis were used to investigate the factors affecting the prognosis of cervical cancer patients.ResultsThe immunohistochemical results showed that the positive rates of AMOTL1 in chronic cervicitis,LSIL,HSIL,and cervical cancer were 12.00%(6/50),17.78%(8/45),28.00%(14/50),and 76.47%(78/102),respectively.The positive expression status of AMOTL1 in chronic cervicitis,LSIL,HSIL and cervical cancer were 12.00%(6/50),17.78%(8/45),28.00%(14/50),76.47%(78/102).The positive rates of YAP in chronic cervicitis,LSIL,HSIL,and cervical cancer were 10.00%(5/50),15.56%(7/45),30.00%(15/50),and 63.73%(65/102),respectively.The positive expression of YAP in cervical cancer tissues was higher than that in other groups(χ^(2)=56.66,P<0.001).The positive rates of TAZ in chronic cervicitis,LSIL,HSIL,and cervical cancer were 8.00%(4/50),17.78%(8/45),30.00%(15/50),and 71.57%(73/102),respectively.Th
