过表达钙调蛋白2基因促进胰腺癌PANC-1细胞增殖、迁移的机制

摘  要:[目的]探讨钙调蛋白2基因对胰腺癌PANC-1细胞生物学行为的影响。[方法]收集2021年6月~2022年4月于医院接受手术治疗的62例胰腺癌患者胰腺癌组织及正常胰腺组织石蜡标本,以实时荧光定量聚合酶链反应行CALM2 mRNA水平检测。将PANC-1细胞分为对照组和过表达组,HPDE细胞作为正常组。检测PANC-1细胞的CALM2 mRNA表达、增殖、凋亡、迁移、侵袭情况和上皮间质转化相关蛋白E-cadherin、Vimentin、N-cadherin及Bcl-2、Bax、caspase3蛋白表达。[结果]CALM2 mRNA在胰腺癌组织中表达水平(2.76±0.31)明显高于癌旁组织(1.32±0.19)(P<0.05)。与正常组(1.06±0.13、0.69±0.06)相比,对照组和过表达组CALM2 mRNA(2.59±0.28、4.25±0.11)及蛋白(0.96±0.11、2.53±0.04)表达上升,与对照组相比,过表达组CALM2 mRNA及蛋白表达上升(P<0.05)。过表达组PANC-1细胞存活率(98.16%±1.29%)、迁移率(88.56±3.33)和侵袭率(93.01%±3.89%)、Bcl-2(1.13±0.18)、Vimentin(1.36±0.10)、N-cadherin(1.39±0.15)蛋白表达水平高于对照组(94.23%±1.01%、76.26±2.89、85.26±3.74、0.89±0.09、1.03±0.12、1.13±0.12);细胞凋亡率(3.16%±0.48%)、Bax(0.23±0.04)、caspase3(0.31±0.04)、E-candherin(0.46±0.09)蛋白表达水平低于对照组(5.26%±0.63%、0.35±0.07、0.43±0.05、0.84±0.13)(P<0.05)。[结论]CALM2在胰腺癌中表达上调,调高CALM2的表达促进PANC-1细胞的增殖、迁移和侵袭,降低PANC-1细胞的凋亡率,促使胰腺癌进展。[Objective]To discuss the biological effect of calmodulin 2 gene overexpression in human pancreatic carcinoma cell line(PANC-1).[Method]The resected and paraffin-embedded specimens of matched cancer tissues and adjacent normal tissues from 62 patients with pancreatic carcinoma who underwent surgical treatment in our hospital from June 2021 to April 2022 were collected.The CALM2 mRNA expression was detected using real-time fluorescence quantitative polymerase chain reaction.PANC-1 cells were assigned to control group and overexpression group,and HPDE cells were used as the normal group.The CALM2 mRNA expression,proliferation,apoptosis,migration and invasion of PANC-1 cells,as well as the expression of epithelial-mesenchymal transition related proteins E-cadherin,Vimentin,N-cadherin and Bcl-2,Bax,caspase3 were measured.[Result]The expression level of CALM2 mRNA in pancreatic cancer tissues(2.76±0.31)was significantly higher than that in paracancerous tissues(1.32±0.19)(P<0.05).Compared with the normal group(1.06±0.13,0.69±0.06),the expression of CALM2 mRNA(2.59±0.28,4.25±0.11)and protein(0.96±0.11,2.53±0.04)increased in the control group and overexpression group.Compared with the control group,the expression of CALM2 mRNA and protein increased in the overexpression group(P<0.05).The survival rate(98.16%±1.29%),migration rate(88.56%±3.33%),invasion rate(93.01%±3.89%),Bcl-2(1.13±0.18),Vimentin(1.36±0.10),N-cadherin(1.39±0.15)protein expression levels in the overexpression group were higher than those in the control group(94.23%±1.01%,76.26±2.89,85.26±3.74,0.89±0.09,1.03±0.12,1.13±0.12);The apoptosis rate(3.16%±0.48%),Bax(0.23±0.04),and caspase3(0.31±0.04)and the protein expression level of E-candherin(0.46±0.09)was lower than that of the control group(5.26%±0.63%,0.35±0.07,0.43±0.05,0.84±0.13)(P<0.05).[Conclusion]The expression of CALM2 is up-regulated in pancreatic cancer.Increasing the expression of CALM2 can promote the proliferation,migration and invasion of PANC-1 cells,reduce the apo
