摘　　要：[目的]探讨PLAC1在肺腺癌中的表达及其对肺腺癌H1299细胞转移的影响。[方法]选择73例肺癌患者和肺腺癌H1299细胞为研究对象,免疫组化法检测肺癌组织及癌旁组织PLAC1表达水平。将肺腺癌H1299细胞分为对照组、空载组、PLAC1 inhibitor组和PLAC1 mimic组。实时荧光定量PCR检测细胞内PLAC1转录水平,蛋白印迹法检测细胞内PLAC1翻译水平,CCK-8试剂检测肺腺癌H1299细胞活力,划痕实验检测肺腺癌H1299细胞迁移率,Transwell法测定肺腺癌H1299细胞侵袭力;流式细胞术检测细胞凋亡率。[结果]肺癌组织中PLAC1表达水平高于癌旁组织(3.82±0.12/分vs 0.96±0.07/分);PLAC1 inhibitor组PLAC1表达水平低于对照组和空载组(0.42±0.03 vs 0.41±0.04 vs 0.12±0.01),PLAC1 mimic组PLAC1表达水平高于对照组和空载组(0.42±0.03 vs 0.41±0.04 vs 0.95±0.11)。与对照组比较,PLAC1 inhibitor组的肺腺癌细胞存活率、迁移以及侵袭能力降低,细胞凋亡率增高,而PLAC1 mimic组肺腺癌细胞存活率、迁移以及侵袭能力增高,细胞凋亡率降低。[结论]肺癌组织中PLAC1表达水平升高,上调PLAC1可促进肺腺癌H1299细胞的增殖和侵袭能力,并抑制期其凋亡。[Objective]To investigate the expression of PLAC1 in lung adenocarcinoma and its effect on the metastasis of lung adenocarcinoma H1299 cells.[Method]Seventy-three cases of lung cancer and lung adenocarcinoma H1299 cells were selected as research objects.The expression of PLAC1 in tissues(lung cancer and adjacent)was testted by immunohistochemistry.Lung adenocarcinoma H1299 cells were divided into control group,empty vector group,PLAC1 inhibitor group and PLAC1 mimic group.Real-time fluorescence quantitative PCR was used to detect the transcription level of PLAC1 in cells,and Western blot was used to detect the translation level of PLAC1 in cells.CCK-8 reagent was used to detect the viability of lung adenocarcinoma H1299 cells,the migration rate and invasiveness of lung adenocarcinoma H1299 cells was detected by scratch test and transwell method.The apoptosis rate was detected by flow cytometry.[Result]The expression level of PLAC1 in lung cancer tissues was higher than adjacent tissues(3.82±0.12/score vs 0.96±0.07/score).The expression level of PLAC1 in PLAC1 inhibitor group was lower than that in control group and empty vector group(0.42±0.03 vs 0.41±0.04 vs 0.12±0.01).The expression level of PLAC1 in the PLAC1 mimic group was higher than that in the control group and the empty vector group(0.42±0.03 vs 0.41±0.04 vs 0.95±0.11).Compared with the control group,the survival rate,migration and invasion ability of lung adenocarcinoma cells in the PLAC1 inhibitor group were decreased,and the apoptosis rate was increased.The survival rate,migration and invasion ability of lung adenocarcinoma cells in the PLAC1 mimic group were increased,and the apoptosis rate was decreased.[Conclusion]The expression of PLAC1 is increased in lung cancer tissues.Up-regulation of PLAC1 could promote the invasion and proliferation of lung adenocarcinoma H1299 cells,and inhibit their apoptosis.
