摘 要:[目的]探讨miR-125-5p靶向调节GCNT1对子宫内膜癌HEC-1A细胞恶性生物学行为的影响。[方法]测定19例子宫内膜癌组织及癌旁组织miR-125-5p、GCNT1水平;设置HEC-1A子宫内膜癌细胞、miR-NC、miR-125-5p过表达、miR-125-5p抑制剂组,测定细胞活力、凋亡、侵袭以及miR-125-5p、GCNT1水平。[结果]癌组织中miR-125-5p水平(1.65±0.82)明显高于癌旁组(0.64±0.18),GCNT1水平(1.45±0.21)显著低于癌旁组(3.93±0.21)(P<0.05)。与HEC-1A组、miR-NC组比较,miR-125-5p过表达组OD值(0.81±0.13)、存活率(82.35%±6.34%)、穿膜数水平(1625.78±23.32)升高,凋亡率(16.30%±1.14%)降低,miR-125-5p mRNA水平升高(3.91±0.14),GCNT1 mRNA(2.17±0.41)、蛋白水平(0.60±0.12)降低(P<0.01);miR-125-5p抑制组OD值(0.37±0.12)、存活率(39.47%±5.41%)、穿膜数水平(402.36±42.24)降低,凋亡率升高(31.40%±1.30%),miR-125-5p mRNA水平降低(0.96±0.13),GCNT1 mRNA(6.37±0.52)、蛋白水平(1.46±0.15)升高(P<0.01);与miR-125-5p过表达组比较,miR-125-5p抑制剂组OD值、存活率、穿膜数水平降低,凋亡率升高,此外miR-125-5p mRNA水平降低,GCNT1 mRNA、蛋白水平升高(P<0.01)。[结论]miR-125-5p在子宫内膜癌组织中高表达,GCNT1在子宫内膜癌组织中低表达,miR-125-5p靶向抑制GCNT1进而促进子宫内膜癌HEC-1A细胞的增殖、侵袭能力,并降低子宫内膜癌HEC-1A细胞的凋亡率。[Objective]To discuss the effect of miR-125-5p-targeted regulation of GCNT1 on the malignant biological behavior of endometrial cancer HEC-1A cells.[Method]The expression levels of miR-125-5p and GCNT1 in paired cancer tissues and adjacent normal tissues from 19 patients were detected.Cells were assigned tothe HEC-1A group,miR-NC group,miR-125-5pmimics group,and miR-125-5pinhibitor group.The cell viability,apoptosis,invasion,and expression levels of miR-125-5p and GCNT1 were determined in each group.[Result]The level of miR-125-5p in cancer tissues(1.65±0.82)was significantly higher than the adjacent tissue group(0.64±0.18),and the level of GCNT1(1.45±0.21)was significantly lower than the adjacent tissue group(3.93±0.21)(P<0.05).Compared with the HEC-1A group and miR-NC group,the miR-125-5p mimicsgroup showed higher OD value(0.81±0.13),survival rate(82.35%±6.34%),and level of number of perforated membranes(1625.78±23.32),and lower apoptosis rate(16.30%±1.14%)(P<0.01),whereas miR-125-5p mRNA levels were increased(3.91±0.14)and GCNT1 mRNA(2.17±0.41)and protein levels(0.60±0.12)were decreased(P<0.01).In the miR-125-5p inhibitor group,OD value(0.37±0.12),survival rate(39.47%±5.41%),level of membrane penetration number(402.36±42.24)were reduced,apoptosis rate was increased(31.40%±1.30%),miR-125-5p mRNA level was reduced(0.96±0.13),GCNT1 mRNA(6.37±0.52),and protein levels(1.46±0.15)were elevated(P<0.01).Compared with the miR-125-5p mimics group,the miR-125-5p inhibitor group had lower levels of OD,survival,and perforation number,and higher apoptosis rates.In addition,miR-125-5p mRNA level was decreased,GCNT1 mRNA and protein levels were increased(P<0.01).[Conclusion]miR-125-5p was highly expressed in endometrial cancer tissues,and GCNT1 was lowly expressed in endometrial cancer tissues.miR-125-5p targeted to inhibit GCNT1 and thus promote the proliferative and invasive ability of endometrial cancer HEC-1A cells,and reduced the apoptosis rate of endometrial cancer HEC-1A cells.
