加味生脉散治疗放射性肠炎的机制研究

摘  要:目的探讨加味生脉散对电离辐射引发的大鼠小肠隐窝上皮细胞(IEC-6)及大鼠肠道损伤的保护作用,并通过NOD样受体热蛋白结构域相关蛋白3(NLRP3)焦亡通路阐明其潜在机制。方法将IEC-6细胞分为空白对照组、模型组、NLRP3沉默组和加味生脉散含药血清组,采用8Gy电离辐射诱导细胞损伤。此外,将大鼠分为空白对照组、模型组和加味生脉散治疗组,给予10 Gy电离辐射建立放射性肠炎模型。通过MTT法、流式细胞术、ELISA和Western blot检测细胞活性、焦亡水平及炎症因子表达,结合大鼠肠道病理学观察,评估加味生脉散的保护作用及其对氧化应激和炎症反应的调控。结果与模型组相比,NLRP3沉默组和加味生脉散含药血清组显著提高了IEC-6细胞的活性,降低了焦亡水平及Caspase-1表达(P<0.01)。在大鼠模型中,加味生脉散显著改善了放射引起的肠道损伤,减轻了辐射引发的氧化应激和炎症反应(P<0.01)。结论加味生脉散通过抑制NLRP3焦亡通路,减轻了电离辐射对IEC-6细胞及大鼠肠道的损伤,展现出抗炎、抗氧化及焦亡抑制的综合作用,为放射性肠炎的预防和治疗提供了新的潜在途径。Objective To investigate the protective effect of Jiawei Shengmai powder on rat small intestine erypt epithelial cells(IEC-6)and rats with intestinal injury induced by ionizing radiation,and to explore its potential mechanism through the NOD-like receptor thermal protein domain associated protein 3(NLRP3)pyroptosis pathway.Methods IEC-6 cells were divided into blank control group,model group,NLRP3 silencing group,and Jiawei Shengmai powder treatment groups,and 8 Gy ionizing radiation was used to induce cell damage.Additionally,rats were divided into control group,model group,and Jiawei Shengmai powder treatment group,where 10 Gy ionizing radiation was used to establish a radiation-induced enteritis model.Then,MTT assay,flow cytometry,ELISA,and Western blot were used to detect cell viability,pyroptosis level,and expression of inflammatory factors.Based on pathological observation of rat intestinal tissue,the protective effect of Jiawei Shengmai powder and its regulatory role in oxidative stress and inflammatory response were evaluated.Results Compared with the model group,the NLRP3 silencing group and Jiawei Shengmai powder treatment group significantly increased the viability of IEC-6 cells,reduced the level of pyroptosis,and decreased the expression of Caspase-1(P<0.01).In the rat model,Jiawei Shengmai powder significantly improved radiation-induced intestinal injury,reduced oxidative stress and inflammatory response(P<0.01).Conclusions Jiawei Shengmai powder mitigates ionizing radiation-induced damage to IEC-6 cells and rat intestines by inhibiting the NLRP3 pyroptosis pathway,exhibiting anti-inflammatory,antioxidant,and pyroptosis-suppressing effects.These findings provide a potential new approach for the prevention and treatment of radiation-induced enteritis.
