摘  要:研究旨在筛选出一株对猪萎缩性鼻炎主要病原菌多杀性巴氏杆菌有良好颉颃作用的产细菌素乳酸菌,分析其细菌素理化特性同时评估其生物安全性。从延边的新鲜蔬菜样品中分离出一株对多杀性巴氏杆菌有良好抑菌活性的乳酸菌,采用形态学观察及16S rDNA序列分析对该乳酸菌进行菌种鉴定并将其命名为MR3;对MR3所产细菌素进行温度、pH、蛋白酶不同单因素处理试验分析其细菌素理化特性。通过药敏试验及全基因组测序对MR3进行生物安全性评估。结果表明:经鉴定MR3为食窦魏斯氏菌,其细菌素在-80~100℃、pH为2.0~10.0时,抑菌活性并未显著下降,表现出良好的稳定性。MR3所产细菌素对大部分蛋白酶表现出耐受性,而对蛋白酶K较为敏感。MR3对常见抗生素均敏感,且未在基因组中发现耐药基因。综上,MR3具有良好的生物安全性,其所分泌的细菌素具有较好的抗菌活性和稳定性,有望成为新型替抗产品应用于猪萎缩性鼻炎的防治。The aim of this study was to screen a bacteriocin-producing lactic acid bacteria with good antago⁃nistic activity against Pasteurella multocida,the main pathogen of swine atrophic rhinitis,and to analyze its bacteriocin physicochemical properties and evaluate its biological safety.A lactic acid strain with a good antagonistic effect against Pasteurella multocida was isolated from fresh vegetable samples in Yanbian,China.The strain was identified by morphological observation and 16S rDNA sequence analysis and named MR3.The physicochemical properties of bacteriocin produced by MR3 were analyzed by different single factor treatment tests of temperature,pH and protease.The biological safety of MR3 was evaluated by drug sensitivity test and whole genome sequencing.The results showed that MR3 was identified as Weissella cibaria,and its bacteriocin activity did not significantly decrease within the range of-80 to 100℃and pH 2.0-10.0,demonstrating good stability.It shows tolerance to most proteases,but is more sensitive to protease K.MR3 was sensitive to common anti⁃biotics,and no drug-resistant genes have been found in the genome.In conclusion,MR3 has good biosafety,and the bacteriocin secreted by MR3 has good antibacterial activity and stability,which is expected to become a new anti-bacterial product for the prevention and treatment of Swine atrophic rhinitis.
