摘　　要：为解决抗生素残留和耐药性问题,研究蛙皮素抗菌肽Caerin2.1体外抑菌活性和稳定性,以期作为抗生素替代物应用于畜禽生产中提供前期基础。利用固相合成法制备蛙皮素抗菌肽Caerin2.1,通过牛津杯法测定蛙皮素抗菌肽Caerin2.1抑菌活性,应用菌落计数法进一步测定最小杀菌浓度(MIC)。并测定不同pH的磷酸-氢氧化钠缓冲液、NaCl缓冲液和温度对蛙皮素抗菌肽Caerin2.1的抑菌率的影响。结果表明:蛙皮素抗菌肽Caerin2.1对革兰氏阴性菌大肠杆菌和金黄色葡萄球菌均表现出明显的抑菌活性,对大肠杆菌的MIC为32μg/mL,对金黄色葡萄球菌的MIC为16μg/mL。分别在pH为4.0、6.0、8.0、10.0的磷酸-氢氧化钠缓冲液,50、100 mmol/L NaCl缓冲液,40、60℃条件下,均表现出较高抑菌活性。蛙皮素抗菌肽Caerin2.1具有较好抑菌活性和稳定性。To solve the problems of antibiotic residues and drug resistance,the in vitro antibacterial activity and stability of AMPs caerin2.1 of Caerin,aiming to establish a preliminary foundation for the application of antibiotic alternatives in livestock production.AMPs caerin2.1 of Caerin was prepared by solid-phase syn⁃thesis method.The antibacterial activity was determined by Oxford cup method,and the minimum bacteri⁃cidal concentration(MIC)was further determined by colony counting method.The effects of different pH values of phosphoric acid-sodium hydroxide buffer,NaCl buffer and temperature on the bacteriostatic rate of AMPs Caerin2.1 of Caerin were determined.It is showed significant antibacterial activity against both gram-negative Escherichia coli and Staphylococcus aureus,with MIC of 32μg/mL against Escherichia coli and 16μg/mL against Staphylococcus aureus.Phosphoric acid-sodium hydroxide buffers with pH values of 4.0,6.0,8.0 and 10.0,50 mmol/L and 100 mmol/L NaCl buffers at 40℃and 60℃showed high bacterio⁃static activity.In conclusion,AMPs caerin2.1 has high antibacterial activity and stability.
