外周T细胞淋巴瘤的铁死亡作用机制及其潜在治疗中药预测

摘  要:目的基于生物信息筛选外周T细胞淋巴瘤(PTCL)铁死亡差异表达基因及其作用机制并预测潜在治疗中药。方法利用GEO数据库GSE132550数据筛选PTCL差异基因并在线进行差异表达分析,利用FerrDb数据库铁死亡相关人类基因,同时筛选共同基因。利用STRING数据库对共同基因进行PPI分析,获取核心基因。利用DAVID数据库对共同基因进行富集分析,利用CoremineMedical数据库对共同基因进行中药预测,使用Autodock Vina软件对关键活性成分进行分子对接验证,并将结果进行可视化呈现。结果分析得到42个PTCL铁死亡差异表达基因,根据筛选条件获得NOX4、GPX2、DUOX1等5个核心基因。富集分析结果提示调节JAK-STAT信号通路、NOD样受体信号通路、Toll样受体信号通路等通路为PTCL铁死亡关键通路。中药预测结果显示主要中药18味,主要为寒性,以柴胡、丹参、鹅不食草等5种中药较为关键,核心活性成分为异鼠李糖、山奈酚、槲皮素。结论柴胡、丹参、鹅不食草等中药所含异鼠李糖、山奈酚、槲皮素等成分通过调控NOX4、GPX2、DUOX1等靶点来调控铁死亡治疗PTCL。Objective To screen the differentially expressed genes with ferroptosis in peripheral T-cell lymphoma(PTCL)using bioinformatics and to predict potential therapeutic herbs.Methods Differential genes of PTCL were screened through the GSE132550 of GEO database and analyzed online.Ferroptosis-related human genes were retrieved from the FerrDb database,with a concurrent screening for common genes.Protein-protein interaction(PPI)analysis was performed on the common genes using the STRING database to identify core genes.Functional enrichment and pathway analysis were conducted on the common genes using the DAVID database.Traditional Chinese medicine predictions were made using the CoremineMedical database,and key active components were verified through molecular docking with AutodockVina software,with results visualized.Results A total of 42 differentially expressed genes related to PTCL and Ferroptosis were identified,and five core genes,including NOX4,GPX2,and DUOX1,were selected based on the screening criteria.Enrichment analysis indicated that the JAK-STAT signaling pathway,NOD-like receptor signaling pathway,and Toll-like receptor signaling pathway were crucial in PTCL Ferroptosis.The traditional Chinese medicine prediction results highlighted 18 types of traditional Chinese medicine,predominantly cold in nature,with five key herbs such as Bupleurum,Salvia miltiorrhiza,and Goosegrass Herb.The core active components were isorhamnose,kaempferol,and quercetin.Conclusions The active components,such as isorhamnose,kaempferol,and quercetin,found in traditional Chinese medicines like Bupleurum,Salvia miltiorrhiza,and Goosegrass Herb,may regulate PTCL by targeting genes including NOX4,GPX2,DUOX1,and others.
