摘  要:目的基于丝裂原活化蛋白激酶(MAPK)信号通路探讨渐进性抗阻训练对慢性肌肉损伤大鼠的修复效果及内在机制。方法将50只SD大鼠分为对照组(n=10)和损伤组(n=40),损伤组用改良离心运动诱导慢性肌肉损伤后再分4组:模型组和低、中、高强度抗阻训练组(每组n=10)。对照组和模型组正常饲养,训练组按不同强度渐进性抗阻训练8周。检测训练前后肌肉力量、耐力,8周后进行组织病理学检查,并采用Western blot检测MAPK信号通路相关蛋白表达。结果在肌肉力量方面,训练4周后低、中、高强度抗阻训练组较模型组提高(P<0.05),8周后各训练组与模型组差异有统计学意义(P<0.01),且高强度组>中强度组>低强度组(P<0.05);肌肉耐力上,4周后低、中、高强度抗阻训练组较模型组延长(P<0.05),8周后各训练组与模型组差异有统计学意义(P<0.01),且高强度组>中强度组>低强度组(P<0.05)。组织病理学方面,对照组大鼠股四头肌肌纤维排列整齐、紧密、大小均匀且无炎症细胞浸润,模型组肌纤维萎缩、变性、排列紊乱且炎症细胞浸润多;低、中、高强度抗阻训练组损伤减轻,肌纤维排列较整齐、炎症细胞浸润减少,高强度组改善效果最佳。与对照组相比,模型组大鼠肌肉组织中(p-ERK1/2)/(ERK1/2)、p-JNK/JNK、p-p38/p38的比值均升高(P<0.05);与模型组相比,低强度、中强度和高强度抗阻训练组大鼠肌肉组织中这些蛋白的磷酸化水平进一步升高(P<0.05),且高强度抗阻训练组升高幅度最大(P<0.01)。结论渐进性抗阻训练可促进慢性肌肉损伤大鼠修复,其可能机制与MAPK信号通路激活有关。Objective To explore the repair effect and underlying mechanism of progressive resistance training on rats with chronic muscle injury based on the MAPK signaling pathway.Methods Fifty Sprague-Dawley(SD)rats were divided into control group(n=10)and injury group(n=40).The injury group was induced with chronic muscle injury by modified eccentric exercise and then further divided into four groups:a model group,and low-intensity,medium-intensity,and high-intensity resistance training group(n=10).The control group and the model group were fed normally,while the training groups underwent progressive resistance training at different intensities for eight weeks.Muscle strength and endurance were measured before and after training.After eight weeks,histopathological examination was carried out,and the expression of proteins related to the MAPK signaling pathway was detected by Western blot.Results In terms of muscle strength,compared with the model group,the muscle strength of the low-intensity,medium-intensity,and high-intensity resistance training group increased after four weeks(P<0.05).After eight weeks,there were significant differences between each training group and the model group(P<0.01),and the muscle strength in the high-intensity group was greater than that in the medium-intensity group,which was greater than that in the low-intensity group(P<0.05).Regarding muscle endurance,compared with the model group,the muscle endurance of the low-intensity,medium-intensity,and high-intensity resistance training group was prolonged after four weeks of training(P<0.05).After eight weeks,there were significant differences between each training group and the model group(P<0.01),and the muscle endurance in the high-intensity group was greater than that in the medium-intensity group,which was greater than that in the low-intensity group(P<0.05).In histopathology,the quadriceps muscle fibers of rats in the control group were arranged neatly,closely,with uniform size,and there was no infiltration of inflammatory cells.In the mode
