摘  要:目的探讨miR-152通过ERBB3/AKT2通路对宫颈癌细胞增殖和侵袭的影响。方法用RT-PCR法检测正常宫颈、低级别病变、高级别病变以及宫颈癌发生发展过程临床组织样本中miR-152的表达情况。用CCK-8法和EDU实验检测HeLa宫颈鳞癌和SiHa宫颈腺癌细胞的增殖活性;Transwell小室检测HeLa细胞侵袭能力。采用免疫印迹方法验证miR-152对ERBB3/AKT2的调控作用。结果与正常宫颈组织相比,宫颈癌组织样本中miR-152相对表达量明显降低(P<0.001)。与转染空载质粒的对照组HeLa与SiHa细胞相比,过表达miR-152后细胞的增殖、侵袭能力明显降低(P<0.01),而敲低miR-152后细胞的增殖、侵袭能力明显增强(P<0.05),而且ERBB3/AKT通路相关蛋白在miR-152过表达组中表达下降,在miR-152敲低组中其表达水平明显升高。结论miR-152可通过调控ERBB3/AKT2通路间接影响宫颈癌的增殖与侵袭。Objective To explore the influence of miR-152 on the proliferation and invasion of cervical cancer cells via the ERBB3/AKT2 pathway.Methods The expression of miR-152 in clinical tissue samples during the development of cervical cancer was detected using the RT-PCR method.The proliferation activities of HeLa cervical squamous carcinoma and SiHa cervical adenocarcinoma cells were examined by the CCK-8 assay and EDU experiment;the invasion ability of HeLa cells was measured by the Transwell chamber.The regulatory effect of miR-152 on ERBB3/AKT2 was verified by Western blotting.Results Compared with normal cervical tissues,the relative expression level of miR-152 in cervical cancer tissue samples significantly decreased(P<0.001).Compared with the control group transfected with the empty plasmid,the proliferation and invasion abilities of cells after overexpression of miR-152 significantly reduced(P<0.01),while the proliferation and invasion abilities of cells after knockdown of miR-152 significantly enhanced(P<0.05).Moreover,the expression of ERBB3/AKT pathway-related proteins decreased in the miR-152 overexpression group while significantly increased in the miR-152 knockdown group.Conclusions MiR-152 can indirectly affect the proliferation and invasion of cervical cancer by regulating the ERBB3/AKT2 pathway.
