摘 要:目的对平顶山市鲁山县的布鲁氏菌进行分型鉴定和体外药物敏感性(药敏)分析,掌握布鲁氏菌病原学特征,为布鲁氏菌病的防控提供依据。方法采集2023年9—12月平顶山市鲁山县人民医院试管凝集试验(SAT)阳性病人(效价>1:100++)的血液样本,用血培养法分离获得的疑似布鲁氏菌,分别用实时荧光定量聚合酶链式反应(PCR)法、血清凝集法鉴定种型和血清型;用全基因组测序法鉴定菌株的多位点序列分型(MLST)型别;用微量肉汤稀释法测定布鲁氏菌对10种抗生素的体外药敏试验。结果19株疑似布鲁氏菌株均为羊种3型、序列型(ST)8型布鲁氏菌;19株布鲁氏菌对左氧氟沙星、利福平、多西环素、链霉素、四环素、环丙沙星、庆大霉素耐药率均为0.00%,对复方新诺明、头孢曲松钠、氨苄西林耐药率分别为31.58%(6/19),47.37%(9/19),94.74%(18/19)。结论鲁山县19株布鲁氏菌是羊种3型、ST8型;对利福平、多西环素、链霉素、左氧氟沙星、四环素、环丙沙星、庆大霉素等药物敏感。Objective To conduct typing and identification,and in vitro drug sensitivity analysis of Brucella strains in Lushan County,Pingdingshan,in order to understand the pathogenic characteristics and provide a scientific basis for the prevention and control of Brucellosis.Methods Blood samples were collected from patients with positive in vitro agglutination test(SAT)results(titer>1:100++)at Lushan County People's Hospital from September to December 2023.Suspected Brucella strains were isolated using blood culture,genotyped via real-time fluorescence quantitative polymerase chain reaction(PCR),serotyped using serum agglutination,and Multi-Locus Sequence Typing(MLST)was performed on whole genome sequences.In vitro susceptibility to12 antibiotics was assessed using the microdilution method with meat broth.Results A total of 19 suspected Brucella strains were classified as sheep type 3 and sequence type(ST)8,All strains showed 0.00%resistance to levofloxacin,rifampicin,doxycycline,streptomycin,tetracycline,ciprofloxacin,and gentamicin.The resistance rates for compound sulfamethoxazole,ceftriaxone sodium,and ampicillin were 31.58%(6/19),47.37%(9/19),and 94.74%(18/19),respectively.Conclusions The 19 Brucella strains were identified as sheep type 3 and ST8 in.Rifampicin,doxycycline,streptomycin,levofloxacin,tetracycline,ciprofloxacin,and gentamicin were found to be effective against there strains.
