摘　　要：目的:探究磷酸甘油酸激酶1(PGK 1)与乳腺癌临床病理参数的关系及其预后意义。方法:(1)通过TCGA、GEO数据库下载乳腺癌与乳腺正常组织中PGK 1 mRNA表达量的矩阵数据和临床数据,分析乳腺癌组织PGK 1 mRNA表达与临床病理特征的关系,采用Logistics回归评估PGK 1 mRNA表达的独立危险因素;使用Kaplan-Meier方法绘制生存曲线分析PGK 1 mRNA表达水平和乳腺癌患者总生存期(OS)的关系。(2)收集2022年1月-2023年1月包头医学院第一附属医院乳腺外科50例乳腺癌及乳腺正常组织标本,分别通过实时荧光定量PCR技术和免疫组织化学染色检测乳腺癌及正常乳腺组织中PGK 1的表达水平,并分析乳腺癌组织PGK 1表达与临床病理参数之间的关系,Logistics回归评估与PGK 1表达的相关独立危险因素。结果:(1)PGK 1mRNA在乳腺癌中的表达水平明显高于正常乳腺组织(P<0.05);(2)ER,PR,HER-2是影响PGK 1 mRNA表达的独立危险因素;(3)PGK 1mRNA高表达与患者总生存期降低呈正相关;(4)PGK 1在乳腺癌中的表达具有统计学意义;(5)ER,PR,HER-2是影响PGK 1表达的独立相关危险因素。结论:(1)PGK 1在乳腺癌中的表达水平显著升高。(2)ER,PR,HER-2与PGK 1表达呈正相关。(3)PGK 1高表达可能是提示乳腺癌不良预后的潜在预测因子。Objective:To investigate the relationship between phosphoglycerate kinase 1(PGK1)and clinicopathological parameters of breast cancer and its prognostic significance.Methods:(1)The matrix data and clinical data of PGK 1 mRNA expression in breast cancer and normal breast tissues were downloaded from TCGA and GEO databases.The relationship between PGK 1 mRNA expression in breast cancer tissues and clinicopathological features was analyzed.Logistics regression was used to evaluate the independent risk factors of PGK 1 mRNA expression.Kaplan-Meier method was used to draw survival curves to analyze the relationship between PGK 1 mRNA expression level and overall survival(OS)of breast cancer patients.(2)From January 2022 to January 2023,50 cases of breast cancer and normal breast tissue specimens were collected from the Department of Breast Surgery,the First Affiliated Hospital of Baotou Medical College.The expression levels of PGK1 in breast cancer and normal breast tissues were detected by real-time fluorescence quantitative PCR and immunohistochemical staining,respectively.The relationship between PGK 1 expression and clinicopathological parameters in breast cancer tissues was analyzed,and Logistics regression was used to evaluate the independent risk factors related to PGK1 expression.Results:(1)The expression of PGK1 mRNA in breast cancer was significantly higher than that in normal breast tissue(P<0.05).(2)ER,PR and HER-2 were independent risk factors for PGK1 mRNA expression.(3)The high expression of PGK 1 mRNA was positively correlated with the decrease of overall survival.(4)The expression of PGK 1 in breast cancer was statistically significant.(5)ER,PR and HER-2 were independent risk factors affecting the expression of PGK1.Conclusion:(1)The expression level of PGK1 in breast cancer is significantly increased.(2)The expression of ER,PR,HER-2 is positively correlated with the expression of PGK1.(3)High expression of PGK1 may be a potential predictor of poor prognosis of breast c ancer.
