摘 要:目的采用超高效液相色谱-四极杆-静电场轨道阱质谱(UPLC-Q-Exactive-Orbitrap-MS)技术对竹叶饮中的化学成分进行研究。方法采用Waters HSS T3 UPLC C_(18)色谱柱(1.7μm,2.1 mm×150 mm),以乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,柱温40℃,流速为0.3 mL·min^(-1),进样量为5μL。质谱采用ESI源,在正、负离子两种模式下采集数据。结合相关文献及化合物裂解规律,比对各化合物的色谱保留时间、母离子质荷比和特征碎片等信息,鉴定竹叶饮的主要化学成分。结果利用UPLC-Q-Exactive-Orbitrap-MS技术,从竹叶饮中分离鉴定了92个化学成分,包括黄酮类26个,有机酸类23个,三萜类10个,倍半萜类13个,香豆素类8个,生物碱类4个,氨基酸类2个,蒽醌类1个,其他类5个。结论首次采用UPLC-Q-Exactive-Orbitrap-MS技术研究竹叶饮中的化学成分,该结果为其质量控制以及进一步的药效物质基础研究奠定了基础。Objective To qualitatively analyze the chemical components of Zhuyeyin by UPLC-Q-Exactive-Orbitrap-MS.Methods Waters HSS T3 UPLC C_(18) column(1.7μm,2.1 mm×150 mm)was used,with gradient elution.Acetonitrile-0.1%formic acid aqueous solution was used as the mobile phase.The column temperature was 40℃,the flow rate was 0.3 mL·min^(-1),and the injection volume was 5μL.ESI sources were used to collect data in both positive and negative ion modes.Based on relevant literatures and compound fragmentation patterns,the chromatographic retention time,parent ion mass to charge ratio,and characteristic fragment information of each compound were compared to identify the main chemical components of Zhuyeyin.Results Totally 92 chemical components were identified,including 26 flavonoids,23 organic acids,10 triterpenes,13 sesquiterpenes,8 coumarins,4 alkaloids,2 amino acids,1 anthraquinone,and 5 others.Conclusion UPLC-Q-Exactive-Orbitrap-MS technology for the first time has identified the chemical components in Zhuyeyin,laying the foundation for its quality control and therapeutic material basis.
