摘 要:目的:癌症恶病质以脂肪萎缩、肌肉丢失、高消耗为主要特征,研究旨在探究高强度间歇运动(high-intensity interval training,HIIT)和中等强度耐力运动(moderate-intensity endurance training,MET)对小鼠脂肪恶病质的影响及其机制。方法:将3周龄C57BL/6雄性小鼠随机分为安静对照组(NC组)、二乙基亚硝胺(diethylnitrosamine,DEN)腹腔注射组(DEN组)、DEN腹腔注射+MET组(MET组)、DEN腹腔注射+HIIT组(HIIT组)。DEN组、MET组和HIIT组小鼠腹腔注射DEN(25 mg/kg,1次/周,共10次),NC组小鼠腹腔注射生理盐水作为对照。随后MET组和HIIT组小鼠进行16周的运动干预,并监测其体成分变化。运动干预结束后采用小动物监测系统监测小鼠能量代谢,采用组织化学染色分析小鼠肝脏、脂肪以及骨骼肌等组织形态改变,采用比色法检测小鼠血清乳酸含量,采用质谱法检测腹股沟白色脂肪组织(inguinal white adipose tissue,iWAT)乳酸水平,采用Simple Western检测iWAT中UCP1、Dio2、Cidea、NDUFS1、PRDM16、MCT1、MCT4、CD147、GPR81和蛋白质泛乳酸化等蛋白表达水平,采用Real-time PCR检测iWAT中Ucp1、Dio2、Cidea、Prdm16等基因的mRNA水平。随后构建运动后乳酸胁迫模型,将3周龄C57BL/6雄性小鼠随机分为DEN腹腔注射组(DIP组),DEN腹腔注射+运动组(ET组),DEN腹腔注射+运动+乳酸输入组(ETL组),各组小鼠腹腔注射DEN,9周龄时进行17周运动干预。ETL组小鼠运动后腹腔注射乳酸钠溶液(2 g/kg,3次/周,共15周)。干预结束后,检测小鼠体成分,采用ELISA检测小鼠血清AFP-L1、TNFα、IL-1β和IL-10的含量。结果:1)与NC组相比,DEN组小鼠体重、体脂率、iWAT和附睾白色脂肪组织(epididymal white adipose tissue,eWAT)质量显著降低,肝指数、肌肉率、日间VVO_(2)以及日间、夜间、总体VVCO_(2)显著增加。DEN组肝肿瘤发生率为45%,MET组肝肿瘤发生率为64%,HIIT组肝肿瘤发生率最低(17%)。2)与DEN组相比,MET组和HIIT组小鼠体Objective:Cancer cachexia is mainly characterized by lipoatrophy,muscle loss,and hypermetabolism.This paper aims to investigate the effects and underlying mechanisms of high-intensity interval training(HIIT)or moderate-intensity endurance training(MET)on adipose cachexia in mice.Methods:3-week old C57BL/6 male mice were randomly divided into four groups:sedentary control group(NC),diethylnitrosamine(DEN)intraperitoneal injection group(DEN),DEN intraperitoneal injection+MET group(MET),and DEN intraperitoneal injection+HIIT group(HIIT).Mice in the DEN,MET,and HIIT groups received DEN(25 mg/kg,once a week for a total of 10 weeks)via by intraperitoneal injection,while mice in the NC group were intra-peritoneally injected with saline as a control.Mice in the MET and HIIT groups underwent 16 weeks of exercise interventions,respectively,with monitoring of changes in body composition.Post-in-tervention,energy metabolism was monitored by using a small animal monitoring system.Histo-chemical staining was used to analyze morphological changes in liver,adipose,and skeletal muscle tissues.Serum lactate levels were detected by colorimetric methods,detection of ingui-nal white adipose tissue(iWAT)in region using mass spectrometry.Protein expression levels of UCP1,Dio2,Cidea,NDUFS1,PRDM16,MCT1,MCT4,CD147,GPR81,and protein lacty-lation in iWAT were detected by Simple Western.mRNA levels of Ucp1,Dio2,Cidea,and Prdm16 in iWAT were detected by Real-time PCR.Subsequently,a post-exercise lactate stress model was established,and 3-week old C57BL/6 male mice were randomly assigned to DEN in-traperitoneal injection group(DIP),DEN intraperitoneal injection+exercise group(ET),and DEN intraperitoneal injection+exercise+lactate infusion groups(ETL).DEN was adminis-tered intraperitoneally to all groups,followed by a 17-week exercise intervention at 9 weeks of age.In the ETL group,mice were intraperitoneally injected with lactate sodium solution(2 g/kg,3 times a week for a total of 15 weeks)after exercise.After the intervention,body compositio
