摘 要:呕吐毒素(DON)广泛存在于自然界,容易污染谷物和饲料,人和动物一旦摄入被污染的谷物和饲料将会对健康造成严重威胁,因此DON的脱毒研究迫在眉睫。本研究针对前期筛选分离得到的一株对低浓度DON有高效降解能力的菌株JM-3,通过形态学显微观察、生理生化反应、全基因组测序和系统发育进化树的构建对其进行分类鉴定;通过绘制其生长曲线了解其生长特征;通过高效液谱法(HPLC)检测JM-3菌株在不同温度、pH和作用时间下对DON的降解效率。结果显示:该菌株的最佳作用条件为:温度30℃、pH 7.0,在48 h内可将DON降解97.8%。该试验为研制德沃斯氏菌JM-3菌株的微生态制剂及进一步应用奠定了基础。Vomitoxin is widely distributed in nature and easily contaminates grains and feed.Once humans and animals ingests contaminated grains and feed,it will pose a serious threat to their health.In this study,a strain JM-3 was isolated before,which was with high efficiency in degrading low concentrations of deoxynivalenol(DON).Classification and identification were performed by screening and identified by morphological microscopic observation,physiological and biochemical reactions,whole genome sequencing,and phylogenetic tree construction;Its growth characteristics were analyzed by drawing its growth curve.The degradation efficiency of DON by JM-3 strain at different temperatures,pH and reaction time were detected by high performance liquid chromatograph(HPLC),and then the optimal action conditions of JM-3 strain were obtained.The results showed that the JM-3 strain was identified as Devosia Sp.,and its optimum reaction conditions were:temperature 30℃,pH 7.0.Under such reaction condition,97.8%of the DON could be degraded within 48 h.Thereafter,the degradation rate of DON was maintained at this level with the time prolonging,which would lay a foundation for the development of microecological agents and further application of the JM-3 strain.
