摘  要:目的探讨枸杞多糖(LBP)对非酒精性脂肪肝大鼠肝脏纤维化和肝脏炎症的作用及机制。方法15只SD大鼠随机分成对照组、模型组和LBP干预组。对照组大鼠正常饲料喂养;模型组大鼠高脂饲料喂养8周;LBP干预组大鼠高脂饲料喂养,每天LBP 10 mg/kg灌胃,持续8周。通过称量大鼠体重、大鼠肝脏湿重,计算肝脏指数。通过RT-qPCR检测各组大鼠肝脏纤维化指标I型胶原蛋白(ColⅠ)、Ⅲ型胶原蛋白(Col-Ⅲ)和α平滑肌肌动蛋白(α-SMA)的表达水平。通过RT-qPCR检测各组大鼠肝脏炎症因子白细胞介素1β(IL 1β)、转化生长因子β1(TGF-β1)以及白细胞介素6(IL-6)mRNA的变化。结果与对照组相比,模型组大鼠肝脏湿重显著降低(P<0.05);肝脏纤维化指标Col-Ⅰ、Col-Ⅲ和α-SMA mRNA的表达水平显著上升(P<0.05);血清中的炎性细胞因子IL-1β、TGF-β1和IL-6 mRNA的表达水平显著上升(P<0.05)。LBP干预组大鼠肝脏湿重显著降低(P<0.05),肝脏纤维化指标Col-Ⅰ、Col-Ⅲ和α-SMA mRNA的表达水平显著上升;血清中的炎性细胞因子IL-1β、TGF-β1和IL-6 mRNA的表达水平显著上升(P<0.05)。与模型组相比,LBP干预后大鼠肝脏湿重显著降低;肝脏中纤维化指标Col-Ⅰ、Col-Ⅲ和α-SMA mRNA的表达水平显著降低(P<0.05);血清中的炎性细胞因子IL-1β、TGF-β1和IL-6 mRNA的表达水平显著降低(P<0.05)。结论LBP能够缓解非酒精性脂肪肝大鼠肝脏纤维化和炎症反应。Objective To explore the effects and mechanisms of lycium barbarum polysaccharide(LBP)on liver fibrosis and liver inflammation in rats with non-alcoholic fatty liver disease.Methods 15 SD rats were randomly divided into control group,model group and LBP intervention group.The rats in the control group were fed with normal feed,the rats in the model group were fed with a high-fat diet for eight weeks,and the rats in the LBP intervention group were fed with a high-fat diet and orally administered 10 mg/kg LBP daily for eight weeks.The liver index was calculated by weighing the rat body weight and rat liver wet weight.The expression levels of liver fibrosis indicators type I collagen(Col-Ⅰ),typeⅢcollagen(Col-Ⅲ)andα-smooth muscle actin(α-SMA)in rats in each group were detected by RT-qPCR.The changes of liver inflammatory factors interleukin-1β(IL-1β),transforming growth factor-β1(TGF-β1)and interleukin-6(IL-6)of rats in each group were detected by RT-qPCR.Results Compared with the control group,the liver wet weight of rats in the model group was significantly reduced(P<0.05).The expression levels of liver fibrosis indicators Col-Ⅰ,Col-Ⅲandα-SMA increased significantly(P<0.05).The serum levels of inflammatory cytokines IL-1β,TGF-β1 and IL-6 increased significantly(P<0.05).The liver wet weight of rats in the LBP intervention group was also significantly reduced(P<0.05).The expression levels of liver fibrosis indicators Col-Ⅰ,Col-Ⅲandα-SMA increased significantly,the levels of inflammatory cytokines IL-1β,TGF-β1 and IL-6 in serum increased significantly(P<0.05).Compared with the model group,the liver wet weight of rats in the LBP intervention group was significantly reduced,the expression levels of fibrosis indicators Col-Ⅰ,Col-Ⅲandα-SMA in the liver were significantly reduced(P<0.05),the levels of inflammatorycytokines IL-1β,TGF-β1 and IL-6 in serum were significantly reduced(P<0.05).Conclusion LBP can alleviate liver fibrosis and inflammatory response in rats with non-alcoholic fatty l
