摘　　要：目的:分析Krüppel样因子5(Krüppel-like factor 5,KLF5)在急性髓系白血病(acute myeloid leukemia,AML)患者及细胞系中的表达水平,探究KLF5在AML患者中的表达对预后和AML细胞生长的影响。方法:检测初诊AML患者及供体骨髓中KLF5的表达情况,分析KLF5表达与患者临床特征及预后的关系。根据GenBank中KLF5的基因序列分别构建过表达KLF5质粒和对照质粒MV4-11/KLF5和MV4-11/Ctrl;借助Real-time PCR验证AML细胞系中过表达KLF5的效果;采用CCK8和细胞克隆形成实验研究KLF5对AML细胞增殖能力的影响。结果:(1)Real-time PCR检测36例AML患者及18例正常供体骨髓样本,结果显示AML患者KLF5 mRNA水平较低;(2)依据KLF5 mRNA表达水平,以中位数为界,将患者分为KLF5低表达组和KLF5高表达组,KLF5高表达组患者3年总体生存(overall survival,OS)率显著高于KLF5低表达组;(3)AML细胞系中KLF5 mRNA表达水平存在差异,KLF5在MV4-11细胞系中的表达水平相对偏低;(4)CCK8法检测结果显示,过表达KLF5基因使MV4-11细胞的增殖速度变慢,克隆形成实验发现过表达KLF5基因使MV4-11细胞克隆形成能力受到抑制。结论:KLF5在AML患者骨髓中表达水平显著低于正常骨髓细胞,且与AML患者的不良预后相关。KLF5对AML细胞增殖和集落形成能力均有抑制作用。Objective:To analyze the expression level of Krüppel-like factor 5(KLF5)in patients with acute myeloid leukemia(AML)and cell lines,and to explore the expression,prognosis and effect of KLF5 on the growth of AML cells.Methods:The expression of KLF5 in bone marrow of newly diagnosed AML patients and donors was detected,and the relationship between KLF5 expression and clinical features and prognosis of patients was analyzed.The expression of KLF5 in AML cell lines was further analyzed,and the overexpressed KLF5 plasmid and control plasmid MV4-11/KLF5 and MV4-11/Ctrl were constructed according to the KLF5 gene sequence in GenBank.Real-time PCR was used to verify the effect of overexpression of KLF5 in AML cell lines.The effect of KLF5 on the proliferation of AML cells was studied by CCK8 and cell clonal formation assay.Results:(1)Real-time PCR was used to detect bone marrow samples from 36 AML patients and 18 normal donors,and the KLF5 mRNA level in AML patients was low.(2)According to the KLF5 mRNA expression level,the patients were divided into low KLF5 expression group and high KLF5 expression group by median.The 3-year overall survival(OS)rate of patients with high KLF5 expression group was significantly higher than that of patients with low KLF5 expression group.(3)The expression level of KLF5 mRNA was different in AML cell lines,and the expression level of KLF5 was relatively low in MV4-11 cell lines.(4)CCK8 assay showed that overexpression of KLF5 gene slowed down the proliferation rate of MV4-11 cells,and clone formation assay showed that overexpression of KLF5 gene inhibited the clonal formation ability of MV4-11 cells.Conclusion:The expression level of KLF5 in bone marrow of AML patients is significantly lower than that of normal bone marrow cells,and it is associated with poor prognosis of AML patients.KLF5 can inhibit the proliferation and colony-forming ability of AML cells.
