摘 要:试验以油茶叶为原料,利用甲醇作为提取溶剂,探究油茶叶总三萜的提取工艺及抗氧化活性。以液料比、提取时间、提取温度、甲醇体积浓度为试验因素,运用响应面法优化油茶叶总三萜提取工艺参数,并对其体外抗氧化活性进行分析。结果显示,在油茶叶总三萜提取过程中,影响提取率的因素按主次顺序为:提取时间、液料比、甲醇体积浓度以及提取温度;最优的提取条件为提取时间2 h、液料比25∶1 mL/g、甲醇体积浓度80%、提取温度65℃,在此条件下油茶叶总三萜化合物提取得率可达0.523%。抗氧化活性研究表明,在一定浓度范围内,油茶叶总三萜提取物对DPPH自由基的清除能力弱于抗坏血酸;但当浓度大于4 mg/mL时,两者清除DPPH自由基能力相当,说明当油茶叶总三萜提取物添加到一定量的时候,具有很强的清除DPPH自由基能力。结果表明:油茶叶总三萜具有较强的抗氧化活性,可用作饲料添加剂、食品、化妆品及医药品原料。The extraction process and antioxidant activity of total triterpenoids from camellia oleifera were studied by using methanol as extraction solvent.Taking the ratio of liquid to material,extraction time,extrac-tion temperature and methanol volume concentration as experimental factors,the extraction process param-eters of total triterpenoids from camellia oleifera abel leaves were optimized by response surface methodol-ogy,and its antioxidant activity in vitro was analyzed.The results showed that in the process of extracting total triterpenoids from camellia oleifera abel leaves,the factors affecting the extraction rate were:extraction time,volume concentration of methanol to liquid and extraction temperature;The optimum extraction condi-tions were as follows:the extraction time was 2 h,the ratio of liquid to material was 25∶1 mL/g,and the volume concentration of methanol was 80%,the extraction temperature was 65℃.Under these conditions,the extraction yield of total triterpenoids from camellia oleifera abel leaves was 0.523%.The study on anti-oxidant activity showed that in a certain concentration range,the scavenging ability of total triterpenoid extracts from camellia oleifera abel leaves was weaker than that of ascorbic acid.However,when the concen-tration is greater than 4 mg/mL,the scavenging ability of both of them is similar,indicating that when the total triterpenoid extract of camellia oleifera abel leaves is added to a certain amount,it has a strong scaveng-ing ability of DPPH free radicals.The results show that the total triterpenoids of camellia oleifera abel leaves have strong antioxidant activity and can be used as feed additives,food,cosmetics and pharmaceutical raw materials.
