摘  要:为优化超声波辅助乙醇法提取杜仲叶中总黄酮的工艺条件,研究测定了不同液料比、乙醇体积分数、超声功率以及超声时间条件下的杜仲叶总黄酮得率,获得最佳单因素条件分别为:乙醇体积分数60%,液料比为30∶1(mL/g),超声功率为540 W,超声时间为30 min。在此基础上,采用响应面试验设计进一步优化提取条件。结果显示,液料比31.82∶1(mL/g)、乙醇体积分数59.84%、超声时间31.14 min、超声功率537.50 W的组合条件下,杜仲叶总黄酮的得率为5.213%;验证试验杜仲叶总黄酮得率实际为5.446%。研究表明,采用上述条件进行超声波辅助乙醇法提取杜仲叶总黄酮得率较高,优化的方法合理可行。In order to improve the total flavonoids yield via ultrasonic-assisted ethanol method,the total fla-vonoids from Eucommia ulmoides under different conditions,such as ethanol concentration,liquid-solid ratio,ultrasonic power and ultrasonic length was tested.The results showed that the best ethanol concentra-tion was 60%,the best liquid-solid ratio was 30∶1(mL/g),the best ultrasonic power was 540 W,and the best ultrasonic length was 30 min.On the above basis,response surface method was further used and obtained the comprehensive conditions consisted of 31.82∶1(mL/g)liquid to solid ratio,59.84%ethanol concentration,31.14 min ultrasonic length and 537.50 W ultrasonic power,then the yield of total flavonoids was 5.213%,while the value becomes 5.446%under the verified optimal conditions.The results illustrated that the total flavonoids receiving rate was increased by ultrasonic and ethanol method under the conditions above,and the method of optimization was reasonable and feasible.
